Abstract
¡¾Abstract Objective? To prepare non-cytotoxic porcine acellular dermal matrix (PADM), evaluate its biological safety, and observe the outcome of the matrix in vivo. method? Take a 0.3-0.4mm thick porcine skin slice, soak it in hypertonic saline to remove the epidermis, and then place it in a sodium hydroxide solution. The dermal cells are removed by continuous shaking in a water bath at room temperature to obtain PADM. After cross-linking and disinfection, bacteriology is performed. and microbiological detection of swine viruses. PADM was implanted subcutaneously in SD rats, and the gross and histological morphological changes of PADM were dynamically observed after surgery. result? The obtained PADM is soft and elastic, easy to shape, and convenient for surgical operations. There are no cellular components in PADM. The collagen fibers are arranged in a regular and orderly manner and the elastic fibers are abundant. Microbiological testing for bacteria and viruses was negative. PADM did not cause immune rejection after being implanted subcutaneously in SD rats. Three weeks after surgery, PADM adhered firmly to the wound and was not easy to peel off from the wound; its structure was still visible 24 weeks after surgery; early PADM was infiltrated by a large number of inflammatory cells, and as time went by, fibroblasts gradually increased and new capillaries appeared. , the collagen fibers gradually become more regular and dense. in conclusion?
