Abstract
¡¾Objective¡¿To isolate and culture goat oviduct epithelial cells and study their growth characteristics. [Method] Use trypsin, collagenase and mechanical scraping methods to isolate fallopian tube epithelial cells and analyze the culture effect; choose immunocytochemical staining to identify the cells; add hypothalamic crude extract, insulin, egf and serum to the culture medium. To explore its effect on the in vitro culture of fallopian tube epithelial cells. [Results] The separation effect of trypsin on goat fallopian tube epithelial cells was poor, followed by collagenase, and the mechanical scraping method was better. Most of the cells isolated by mechanical scraping were cell clusters. They began to grow adherently 8 to 12 hours after inoculation, and the cells took on an obvious paving stone shape after 2 to 3 days of growth. Immunocytochemical staining showed that the positive rate of cytokeratin was over 95%. The epithelial cell culture system contains a small amount of serum, human EGF, and insulin, which can effectively promote the growth of primary and passaged fallopian tube epithelial cells. The culture effect is better with a volume fraction of 5% serum + EGF. [Conclusion] Fallopian tube epithelial cells isolated and cultured by mechanical scraping can be continuously passed down to the 8th passage in vitro. The passage cells have strong activity and high purity, which can provide materials for research on the related functions of fallopian tube epithelial cells.
