Abstract
Phosphoric acid pretreatment was used to assist in the separation of tilapia skin dermis and epidermis, and low-temperature acetic acid swelling method was used to extract collagen in the dermis, and the physical and chemical properties of the obtained collagen samples were analyzed. The results show that the best separation effect of dermis and epidermis can be obtained by treating fish skin with 0.006 mol/L phosphoric acid and 50 g/L liquid for 3 hours in a 4 ¡æ environment. The obtained dermis was swollen with a 0.035 mol/L acetic acid solution (mass concentration of the material liquid was 100 g/L) for 6 hours at 4¡ãC. After extrusion, a dermal collagen sample with a mass fraction of hydroxyproline of 7.9% was obtained. , the extraction yield reaches 27.6% (dry basis). Analysis of the gel electrophoresis results showed that the obtained sample mainly contained ¦Á1, ¦Á2 and ¦Â chains. The UV spectrum analysis results showed that its maximum absorption was near the wavelength of 230 nm, which was consistent with the characteristics of typical type I collagen; attenuated total reflection Fourier transform infrared spectrum The analysis results show that its triple helix structure is well preserved. The thermal denaturation temperature and heat shrinkage temperature of the sample are 29.2 ¡æ and 55.0 ¡æ respectively. The research results provide a theoretical reference for the efficient development and utilization of fish skin resources and the preparation of high-quality collagen.
