Changes In Epithelial-Mesenchymal Phenotype Of Liver In Rats With Liver Fibrosis Induced By Bile Duct Ligation

Abstract

Abstract: Objective To observe whether epithelial-mesenchymal phenotype changes occur in hepatocytes of rats with liver fibrosis induced by bile duct ligation. Methods 24 wistar rats were divided into sham operation group and bile duct ligation group. Hepatic fibrosis and massion staining were used to detect changes in liver fibrosis. Epithelial markers e-calcin, albumin, type I collagen, and waveform in rat liver were detected by immunoblotting. Protein expression; immunofluorescence double staining method was used to detect fibroblast-specific protein (fsp-1) + vimentin; fsp-1 + type I collagen; fsp-1 + albumin; albumin + type I collagen and other proteins localization in cells. Results The ishak fibrosis score (4.42¡À1.16vs0.00¡À0.00, p=0.000), metavir fibrosis score (3.42¡À0.67vs0.00¡À0.00, p=0.000) and collagen content in massion staining of rats in the bile duct ligation group (0.30¡À0.06vs0.11¡À0.07, p=0.000) was significantly higher than that of the sham operation group. Compared with the sham operation group, the epithelial markers albumin (0.53¡À0.63vs1.12¡À0.01, p=0.000) and e-calcin (0.21¡À0.01vs0.44¡À0.01, p=0.000) were significantly reduced in the bile duct ligation group. , while type I collagen (8.21¡À0.12vs0.24¡À0.01, p=0.000) and vimentin (3.14¡À0.01vs0.37¡À0.01, p=0.000) were significantly increased. In the bile duct ligation group, the number of cells co-expressing fsp-1+vimentin, fsp-1+type I collagen, fsp-1+albumin, and albumin+type I collagen was significantly increased compared with the sham operation group. Conclusion In the livers of rats with liver fibrosis induced by bile duct ligation, epithelial markers decreased and mesenchymal markers increased, suggesting that some epithelial cells may have undergone epithelial-mesenchymal phenotypic transition.
