Abstract
In order to determine the main signaling pathways regulated by FGF2 in dairy cow mammary gland branches, adolescent cow mammary gland organoids were first embedded in rat tail type I collagen and added FGF2 for three-dimensional culture; through FGFR1 receptor inhibitor (FGF2+PD173074 group), PI3K/AKT signal Pathway inhibitor (FGF2+LY-294002 group) or MAPK/ERK signaling pathway inhibitor (FGF2+U0126 group) blocked the signaling pathway to determine the mammary gland branching rate of cows under different treatments at 84 h; Western blot was used to detect treatment time 0 and Relative protein expression of p-AKT, AKT, p-ERK and ERK in each group at 15, 60, 240 and 600 min. Studies have found that FGF2 mainly affects dairy cow mammary gland branches through the MAPK signaling pathway. This article reveals the downstream signaling pathway of FGF2 that plays a major role in affecting the branching morphogenesis of the mammary gland in dairy cows, and promotes research on the molecular mechanisms of mammary gland branching morphogenesis in dairy cows.
