Abstract
The cytoskeletal system is the gravity sensing system within the cell. It is known that various changes in cell morphology, function, and signal transduction caused by microgravity are related to changes in the cytoskeleton system, but little is known about the impact of microgravity on related gene regulation. This study took the constructed genetically engineered cell line (EGFP–ROS) as the object, used a gyrator to simulate the microgravity effect, and used enhanced green fluorescent protein (EGFP) fluorescence semi-quantitative and cell microfilament fluorescence staining analysis. technology to explore the effect of the cell microfilament system on the promoter activity of the collagen type I alpha chain 1 gene ({sl COL1A1}) under rotational simulated microgravity conditions. After space flight and rotation simulated microgravity, cell microfilaments depolymerized and tension fibers decreased, indicating that microgravity can reduce the orderliness of cell microfilament structure and induce cytoskeletal rearrangement. Treatment of EGFP-ROS cells with an appropriate dose of cytochalasin B induced the depolymerization of the microfilament skeleton, which also led to an increase in {sl COL1A1} promoter activity and enhanced cell fluorescence intensity in a dose-dependent manner. Therefore, a certain degree of disruption of the cell microfilament system will lead to the enhancement of {sl COL1A1} promoter activity, proving that the cell microfilament skeleton system is involved in the regulation of {sl COL1A1} promoter activity by microgravity and plays a role in microgravity signal transduction. important role.
