Abstract
Objective To explore the use of differentiated endothelial cells induced by bone marrow stromal stem cells (BMSCs) as seed cells, using collagen-chitosan complex as scaffold material, and using the principles and methods of tissue engineering to construct tissue engineering heart valves (TEHV) in vitro ) feasibility. Methods: Smooth muscle cells and fibroblasts were isolated, cultured and passaged from the rabbit aortic wall in vitro. The cultured fibroblasts, smooth muscle cells and endothelial cells differentiated from rabbit bone marrow stromal stem cells were sequentially planted on pre-wetted collagen-chitosan. On the composite scaffold, the growth, proliferation and matrix secretion of endothelial cells, fibroblasts and smooth muscle cells on the scaffold were observed through histology, inverted microscope and scanning electron microscope. Results HE staining showed that endothelial cells could adhere, stretch, and proliferate on the collagen-chitosan composite scaffold, and grew into a continuous cell layer on the TEHV surface; scanning electron microscopy showed that the TEHV surface was smooth, the cells grew well, the cell layer was intact, and the valve surface Endothelial cells have the function of synthesizing and secreting prostacyclin (PGI2). Conclusion TEHV can be constructed by using the smooth muscle cells, fibroblasts and endothelial cells induced and differentiated by BMSCs isolated, cultured and passaged from the rabbit aorta wall in vitro as seed cells and then planted on the collagen-chitosan composite scaffold, and the endothelial cells can secrete PGI2.
