Therapeutic Effect Of Fuzheng Huayu Prescription Combined With Huangqi Decoction On Rats With Experimental Liver Cirrhosis

Abstract
Objective: To observe the therapeutic effect of Fuzheng Huayu prescription combined with Huangqi decoction on rats with experimental cirrhosis, and to explore the characteristics of the therapeutic effect of deficiency-tonifying traditional Chinese medicine on liver cirrhosis. Methods: A rat model of liver cirrhosis was prepared by intraperitoneal injection of dimethylnitrosamine (DMN) for 4 weeks. After the model was formed, the model rats were given Fuzheng Huayu Recipe, Huangqi Decoction, Fuzheng Huayu Recipe and Huangqi Decoction by gavage for 2 weeks. Observe the body weight, liver weight, and spleen weight of rats; HE staining to observe inflammation and pathology of liver tissue; Sirius red staining to observe collagen deposition in liver tissue; The kit detects rat serum liver function (ALT, Alb, T.Bil) and liver tissue SOD activity. and MDA content; Hyp content in liver tissue was determined by hydrochloric acid hydrolysis method; ¦Á-SMA expression in liver tissue was observed by immunohistochemical staining; ¦Á-SMA protein expression in liver tissue was detected by Westernblot. Results: Compared with normal rats, the serum ALT and T.Bil of the model rats were significantly increased, and the Alb content was significantly decreased; the SOD activity of the liver tissue decreased, and the MDA and Hyp contents were significantly increased; the inflammatory lesions and collagen deposition in the liver tissue were obvious. ¦Á-SMA expression was significantly increased. Huangqi decoction, Fuzheng Huayu prescription and their combination can significantly improve the serum liver function of model rats, improve liver lipid peroxidation, reduce liver tissue Hyp content, reduce liver tissue inflammation and collagen deposition, and inhibit ¦Á-SMA expression. In terms of improving liver collagen deposition and inhibiting ¦Á-SMA expression, the combination of the two is better than either alone. Conclusion: Fuzheng Huayu Recipe, Huangqi Decoction, Fuzheng Huayu Recipe and Huangqi Decoction can all improve liver fibrosis in DMN model rats. The combination of the two prescriptions inhibits collagen deposition more significantly than each single prescription group. Part of the mechanism is that the combination of the two prescriptions can improve liver fibrosis. Good at inhibiting hepatic stellate cell activation.
