Abstract
Objective To explore the regulatory effect of angiotensin ¢ò (Ang ¢ò) on the promoter activity of human ¦Á1(¢ñ) collagen gene and the influence of ligustrazine intervention. Methods The recombinant pCOLH 2.5, which consists of a 2.5 kb human ¦Á1(I) collagen gene promoter fragment and a chloramphenicol acetyltransferase (CAT) “reporter gene”, was transfected into cultured rats using liposome method. Cardiac fibroblasts were added with Ang¢ò and ligustrazine, and CAT activity was measured by ELISA. Results Ang¢ò can significantly promote the priming activity of pCOLH 2.5 fragment, and ligustrazine can inhibit the pCOLH 2.5 priming activity of Ang¢ò (P<0.05). Conclusion Ligustrazine can block the Ang II-induced increase in human ¦Á1(I) collagen gene promoter activity at the transcriptional level, thereby inhibiting the synthesis of type I collagen and blocking and delaying the occurrence and development of myocardial fibrosis.
