Abstract

Objective To observe the effect of triptolide on bone and joint destruction in collagen-induced arthritis (CIA) model mice. Methods DBA/1 mice were immunized subcutaneously with type II collagen twice to establish a CIA model. 14 to 42 days after the first immunization, triptolide (8.18, 16.36, 32.72 ?g/kg) was administered to each treatment group. ) and methotrexate (0.1 mg/kg), clinical symptoms and onset were observed regularly, samples were collected on day 42, histopathological and radiological changes were observed, and tartrate-resistant acid phosphatase (TRAP) was used to specifically stain osteoclasts. Then TRAP-positive cells were counted. Results Triptolide can significantly reduce the clinical scores and incidence of CIA mice, reduce the histopathological changes of joint synovium, cartilage and bone and the degree of bone destruction in X-ray films, improve the joint space, and increase the bone density of knee joints and lumbar vertebrae. density, reducing the number of TRAP-positive cells in the bone marrow cavity and inflammatory granulation tissue of the knee joint. Conclusion Triptolide has a certain inhibitory effect on joint inflammation and bone destruction in CIA mice
