Histochemical Characteristics And Ultrastructural Comparison Of Extracellular Matrix-Related Proteins In Bactrian Camel Testis And Cryptorchidism

Abstract
Abstract In order to explore the distribution and histochemical characteristics of extracellular matrix-related proteins in cryptorchid Bactrian camels, electron microscopy and various histochemical methods were used to compare the ultrastructure, histochemical characteristics and laminar adhesions of cryptorchid and normal testes. Distribution characteristics of protein (LN), type IV collagen (Col IV) and heparan sulfate glycoprotein (HSPG). The results show that: (1) Compared with the normal testicular interstitial structure, cryptorchid seminiferous tubules under light microscopy are underdeveloped, collagen fibers in the interstitium are sparse, reticular fibers are obviously distributed, interstitial blood vessels and the membrane proper of seminiferous tubules PAS and AB-PAS positive reaction is weak. Under the electron microscope, the basement membrane of cryptorchid spermatogenic epithelium is significantly proliferated, with fewer peripheral type I collagen fibers and atypical peritubular myoid cells; fibroblasts are more common around interstitial capillaries and Leydig cells, while in normal testes, interstitial capillaries are present. Fibroblasts are mostly distributed around Leydig cells. (2) Immunohistochemistry staining showed that Col IV, LN and HSPG in normal testicular tissue were all strongly positively expressed in Leydig cells, and Col IV and LN were strongly expressed in capillary endothelial cells, with the latter being especially expressed in Sertoli cells. Obviously, HSPG is not expressed in spermatogonia; in cryptorchidism, the positive expression of Col ¢ô, LN and HSPG in Leydig cells is significantly weakened, and the positive expression of Col ¢ô and LN in peritubular myoid cells and capillary endothelial cells is also significantly weakened. HSPG is strongly positively expressed in spermatogonia and sperm cells. Immunohistochemistry image analysis results showed that the distribution of Col IV and LN in the normal testicular tissue of Bactrian camels was significantly higher than that in cryptorchidism tissue (P<0.05). The HSPG test results showed no statistical difference between normal testicles and cryptorchidism (P >0.01). This study shows that the development of seminiferous tubules in Bactrian camel cryptorchidism is abnormal, the ability to synthesize collagen fibers in the interstitial tissue is reduced, and the important components of testicular extracellular matrix, Col IV and LN, are significantly different from the normal group and are related to abnormalities in seminiferous tubules and Leydig cells. It is related to development, and the strong positive expression of HSPG in cryptorchid spermatogenic epithelium is closely related to the immaturity of spermatogonia.
