Abstract
The purification process for preparing high-purity red stingray cartilage angiogenesis inhibitory factor was determined, and its biological activity was verified through collagenase model and CAM model. Orthogonal experiments determined the optimal conditions for extracting red stingray cartilage tissue, and the extraction yield could reach 0.84%. Acetone fractionated precipitation was used to roughly separate the extraction products, and the CAM angiogenesis inhibition rate of 25-35% acetone fractionated products reached 85%. By optimizing the conditions of ion exchange chromatography, gel filtration, and reversed-phase chromatography, high-purity red stingray cartilage angiogenesis inhibitory factor-I (DCAIF-I) can be further effectively obtained. SDS-PAGE-Coomassie brilliant blue staining showed a band with a molecular weight of 62kDa. Activity verification results show that DCAIF-I has a certain inhibitory activity on collagenase, with an inhibitory rate of 36.3%, and its inhibitory activity on CAM angiogenesis has a certain dose-dependent relationship.
