Abstract
Objective To study the role of autocrine collagen type I (collageni) in lung cancer cells in three-dimensional culture of lung cancer cells. Methods: It was confirmed that lung cancer cells express collageni through immunohistochemistry and rt-PCR; the sh-collageni method was used to interfere with the expression of lung cancer cells, and normal lung cancer cells served as the control group; the supernatant of fibroblasts was used to stimulate tumors, and rt-PCR was used to detect the expression of collageni. Express. Results: Lung cancer cells secreted collageni; it was found that the three-dimensional growth of lung cancer cells knocking out collageni was restricted: their growth rate decreased and colony formation in three-dimensional culture was reduced. When fibroblast culture cell supernatant was used to stimulate tumor cells, the expression of autocrine collageni in lung cancer tumor cells changed. Tumor autocrine collageni can maintain its own growth needs and is regulated by the non-extracellular matrix in the microenvironment. Conclusion Lung cancer cells autocrine collageni plays an important role in tumor growth.
