Inhibitory Effect Of Cordyceps Militaris On The Progression Of Renal Disease In Diabetic Rats

Abstract
Objective: To study the inhibitory effect of Cordyceps militaris on the development of renal disease in diabetic rats. Methods: A diabetic rat model was established using a one-time intraperitoneal injection of streptozotocin (STZ). After oral administration of Cordyceps militaris (400 mg/kg, based on body weight) for 8 weeks, the fasting blood glucose levels and renal index of the rats were measured respectively; 24-hour urine of the rats was collected to detect trace albumin (mAlb) and ¦Â in the urine. -N-acetylglucosidase (NAG) content; determination of urea nitrogen (BUN), creatinine (Cr), and uric acid (UA) contents in serum; determination of transforming growth factor ¦Â1 (TGF-¦Â1) and its contents in kidney by RT-PCR method Type II receptor (T¦ÂR¢ò) and connective tissue growth factor (CTGF) mRNA expression; Western blotting to detect TGF-¦Â1 protein expression in the kidney; immunohistochemistry to detect type IV collagen expression in the kidney. Results: Cordyceps militaris can significantly reduce fasting blood glucose (P£¼0.05), renal index (P£¼0.01), 24h urine output (P£¼0.01), urinary microalbumin excretion rate (UAER) (P£¼0.01) and NAG excretion rate (P<0.01); at the same time, it can reduce the levels of BUN, Cr, and UA in serum (P<0.05); down-regulate the mRNA expression of TGF-¦Â1, TGF¦ÂR¢ò and CTGF (P<0.01), and reduce TGF-¦Â1 and type IV Collagen protein expression (P<0.01). Conclusion: Cordyceps militaris has good effects on lowering blood sugar and improving kidney filtration function. Its mechanism may be related to inhibiting the expression of genes such as TGF-¦Â1, TGF¦ÂR¢ò and CTGF, and reducing the accumulation of collagen substances in the kidneys.
