Abstract
Objective To explore the effects of liver fibrosis factors matrix metalloproteinase-8 (MMP-8), matrix metalloproteinase inhibitor-2 (TIMP-2) and type I and type III collagen (Col-I and Col-III) on arsenic exposure in water. Expression and significance in mouse liver tissue. Methods 50 male mice were randomly divided into control group, sodium arsenite group and sodium arsenate group. The sodium arsenite group and the sodium arsenate group were given ordinary feed and arsenic solution. The mice were sacrificed after 10 months, and the serum was used for liver function testing; part of the liver tissue was stained with hematoxylin-eosin (HE); total RNA from the mouse liver tissue was extracted with the Trizol-phenol-chloroform one-step method, and the total RNA concentration and Purity, real-time fluorescence quantitative PCR method was used to measure the expression of MMP-8, MMP-2, Col-¢ñ, and Col-¢ómRNA in liver tissue, and the 18S gene was used as a quality control. Results The serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and globulin (GLB) values in the sodium arsenite group and the sodium arsenate group were all higher than those in the control group; liver tissue pathological examination showed obvious liver Cell necrosis, regeneration and a little fibrosis; compared with the control group, the differences in the expression of Col-¢ñ and Col-¢ómRNA in the liver tissue of the sodium arsenite group were statistically significant (tCol-¢ñ=5.49, tCol-¢ó, P<0.05 ); the difference in MMP-8 mRNA decrease was statistically significant (t=7.35, P<0.05); the differences in TIMP-2, Col-¢ñ, and Col-¢ómRNA in the sodium arsenate group were statistically significant (tTIMP-2=6.72, tCol -¢ñ=549, tCol-¢ó=9.68, P<0.05); the difference in MMP-8mRNA reduction was statistically significant (t=6.28, P<0.05). Conclusion Long-term exposure to arsenic in drinking water can lead to chronic liver damage and liver fibrosis; TIMP-2 and MMP-8 genes play an important role in the occurrence of arsenic-induced fibrosis in mice.
