Abstract
The collagen content in the dermis of mammalian skin is about 70%, mainly type I and type III collagen. This experiment used dilute acid dissolution and enzymatic extraction of total collagen in rat skin, and crude collagen extract After denaturation at 60¡ãC, trypsin was used for degradation. Liquid chromatography/mass spectrometry was used to analyze the characteristic peptides of the two collagens. The characteristic peptides were used to compare the relative contents of type I and type III collagen in the skin of rats at different growth stages. The results showed that the relative content of type III collagen in rat skin gradually decreased with the extension of the growth period, while the relative content of type I collagen gradually increased, and the ratio of the two collagens tended to be stable after 8 weeks. The results of this experiment demonstrate the feasibility of using high-performance liquid chromatography/mass spectrometry to analyze collagen types and dynamic changes in tissues, providing an experimental basis for better clinical applications.
