Angiotensin ¢ò Induces The Synthesis Of Type ¢ñ Collagen In Passively Sensitized Human Airway Smooth Muscle Cells

Abstract
Objective To explore the effect of angiotensin ¢ò (Ang ¢ò) and its type ¢ñ receptor (Angiotensin Type 1 Receptor, AT1R) antagonist Losartan (Losartan) on the synthesis of type ¢ñ collagen in passively sensitized human airway smooth muscle cells (HASMCs). Methods HASMCs were cultured in vitro, and the cells were divided into 4 groups according to treatment factors: ¢Ù Passive sensitization group (10% asthma serum); ¢Ú Passive sensitization + Ang¢ò group (10% asthma serum + 10-7mol/LAng¢ò); ¢Û Passive sensitization group Allergy+Losartan group (10% asthma serum+10-6mol/LLosartan); ¢ÜPassive sensitization+Ang¢ò+Losartan group (10% asthma serum+10-7mol/LAng¢ò+10-6mol/LLosartan). Immunofluorescence staining was used to identify HASMCs, fluorescence quantitative PCR was used to detect type I collagen mRNA expression, and ELISA was used to detect type I collagen secretion. Results After 10-7 mol/l Ang¢ò acted on passively sensitized HASMCs for 24 hours, the expression of type I collagen mRNA and protein increased significantly compared with the passive sensitization group (P<0.01). In the presence of Losartan, the promoting effect of Ang II on the expression of type I collagen mRNA and protein in passively sensitized HASMCs was significantly inhibited (P<0.01). Conclusion Ang¢ò can promote the secretion of type I collagen in passively sensitized HASMCs, possibly by binding to AT1R.
