Objective To observe the anti-tumor effect of anti-type IV collagenase monoclonal antibody 3g11 and lidamycin (ldm) conjugate. Methods: The mtt method was used to determine its inhibitory effect on tumor cell proliferation; mouse transplanted liver cancer h22 was used to observe the anti-tumor effect in vivo. Results The 3g11-ldm conjugate retained the binding ability of monoclonal antibody 3g11 to type IV collagenase and target cells h22 cells. In vitro tests on h22 cells showed a stronger inhibitory effect on cell proliferation than free ldm. The tumor inhibition rates of 3g11-LDM conjugates 0.05 and 0.10 mg¡¤kg-1 in vivo on mouse transplanted liver cancer h22 were 87.8% and 97.2% respectively, while the tumor inhibition rate of free LDM 0.05 mg¡¤kg-1 was 67.1%. , and the median survival time of mice in the 3g11-ldm conjugate group was significantly longer than that in the ldm group. Conclusion The 3g11-ldm conjugate has a stronger anti-tumor effect on mouse transplanted liver cancer h22 than ldm, and may become an anti-tumor targeted drug.
