Antihypertensive Effect Of Collagen Hydrolysates From Different Parts Of Porcine Femur

Abstract
Porcine whole femur, femoral shaft, and femoral head collagen hydrolyzate were used as samples to study their effects on blood pressure in spontaneously hypertensive rats (SHR). After SHR adaptive feeding for 1 week, they were randomly divided into 11 groups, with 8 animals in each group: the negative control group was gavaged with distilled water, and the 3 samples were divided into 3 dose groups: low, medium, and high, respectively, at 20.0, 40.0, and 60.0 mg/kg. The corresponding collagen hydrolyzate was intragastrically administered at a dose of 10.0 mg/kg, and the positive control group was intragastrically administered captopril at a dose of 10.0 mg/kg. The SHR tail artery systolic pressure of each group was measured every 2 hours after intragastric administration. The results showed that the low-dose femoral head group, whole femur and femoral stem sample groups had no significant antihypertensive effect on SHR, while the mid- and high-dose femoral head groups could significantly reduce the blood pressure of SHR. UV spectrophotometry was used to detect the ACE inhibition rate of the samples. The ACE half-inhibitory mass concentrations (IC50) of the whole femur and femoral head samples were 3.546 and 2.045 mg/mL respectively. It can be seen that femoral collagen hydrolyzate can effectively inhibit the activity of ACE in vitro, but only femoral head collagen hydrolyzate can effectively reduce SHR blood pressure under higher mass concentration conditions.
