Abstract
Abstract Purpose: To explore the role of macrophage receptor with collagen-like structure (MARCO) in the process of enhancing bacterial lipoprotein (BLP) resistance to macrophage phagocytosis. Methods: Isolate, differentiate and culture bone marrow-derived macrophages (BMDMs), and prepare BLP-resistant cells; use flow cytometry and fluorescence microscopy to detect and compare the ability of BLP-resistant and non-tolerant cells to phagocytose bacteria. Flow cytometry and qPCR were used to detect the effect of BLP tolerance on MARCO protein and mRNA expression; small interfering RNA was further used to downregulate MARCO expression, and the effect of MARCO on the phagocytosis ability of BLP-resistant macrophages was explored through flow cytometry and fluorescence technology. Results: The amount of phagocytosis of bacteria by BLP-tolerant macrophages was significantly higher than that of non-tolerant macrophages (P<0.05); at the same time, the expression level of MARCO protein on the membrane surface of BLP-tolerant macrophages was significantly higher than that of non-tolerant cells, and in It further increased after bacterial stimulation, and the changes in MARCO's mRNA levels were consistent with changes in protein levels; after down-regulating MARCO expression, the phagocytosis ability of BLP-resistant macrophages significantly decreased, suggesting that the enhanced phagocytosis ability of BLP-resistant macrophages is related to the macrophage membrane.
