Abstract
Abstract: Objective To dynamically observe the changes in cardiac morphology and myocardial collagen in ApoE gene-deficient mice of different ages fed with high cholesterol. Methods ApoE gene-deficient mice (n=6/group) aged 24, 32 and 40 weeks were used as the research subjects, and C57BL/6J mice of the same age were set as controls. Serum cholesterol levels were routinely detected; HE staining was used to observe the morphological changes of the hearts of mice in each group, Masson staining was used to observe myocardial collagen changes, and the KS400 image analysis system was used to measure the myocardial collagen volume fraction (CVF) and the ratio of the collagen area and lumen area around myocardial blood vessels ( PVCA), picric acid Sirius red staining for left ventricular collagen specific staining and quantitative analysis to observe changes in type I and type III collagen. Results The left ventricular anterior wall, left ventricular lateral wall, and left ventricular posterior wall of 40-week-old ApoE-/- mice were thicker than those of 24-week-old ApoE-/- mice (P<0.05); 24-, 32-, and 40-week-old ApoE The collagen content in the myocardium of -/- mice increased with age (P<0.05), and type I collagen gradually increased. Conclusion ApoE gene-deficient mice fed with high cholesterol have an enlarged heart and a significant increase in myocardial collagen content as cholesterol levels increase.
