Catalase Immobilization Using Fe(Iii) Modified Collagen Fiber As Carrier

Abstract
Collagen fibers were modified with ferric iron as a carrier, and catalase was fixed on the carrier through glutaraldehyde cross-linking. The prepared immobilized catalase protein had a loading capacity of 16.7 mg/g and an enzyme activity yield of 35%. The optimal pH, optimal temperature, thermal stability, storage stability and operational stability of the immobilized enzyme and the free enzyme were studied. The results show that after catalase is immobilized by this method, the optimal pH and optimal temperature are the same as those of the free enzyme, which are ph7.0 and 25¡ãC respectively; however, the thermal stability of the immobilized enzyme is significantly improved and can be stored at 75¡ãC. After 5 hours, 30% of the activity was still retained, while the free enzyme was completely inactivated; after the immobilized enzyme was stored at room temperature for 12 days, the enzyme activity still remained above 88%, while the free enzyme was completely inactivated under these conditions.
