Abstract

? Purpose: To initially explore the potential of human amniotic membrane stromal cells (hamc) as seed cells for bone tissue engineering. Methods: hamc was isolated and cultured, and the third generation hamc was induced into osteogenesis in DMEM culture medium containing dexamethasone (0.1 ¦Ìmol/l), vitamin C (50 mg/l) and ¦Â-glycerol sodium phosphate (10 mmol/l). After culturing for 1 week, alizarin red staining was performed to count the number of calcified nodules, and immunofluorescence cytochemical staining was performed to detect the expression of type I collagen and alkaline phosphatase. Immunohistochemical staining was used to detect the expression of fasl in amniotic membrane and hamc. Results: After osteogenic induction of hamc, obvious calcified nodules were formed, with an average of 18 per well. The cells in the calcified nodules expressed positive expression of type I collagen and alkaline phosphatase. The positive expression of fasl can be detected in cells in human amniotic membrane tissue and in hamc cultured in vitro. Conclusion: hamc has the potential to become an ideal seed cell for bone tissue engineering.

Abstract

Objective To explore the effect of miR-144 overexpression lentivirus on the pathological process of synovial tissue in arthritis model mice and its possible mechanism. Methods: Type II collagen emulsion was injected subcutaneously into the tail base of BALB/c mice to construct a mouse model of collagen-induced arthritis (CIA), and then a lentivirus with high expression of miRNA-144 was injected into the tail vein. Then, immunohistochemistry was performed on the synovial tissue of the model mice to detect the expression and distribution of Cadherin-11 and TNF-¦Á, and HE staining was used to detect the pathological changes in the synovial tissue. Results The synovial tissue of the model mice showed similar characteristics to the pathological process of rheumatoid arthritis. The inflammatory response of the synovial tissue of the model mice treated with lentivirus with high expression of miRNA-144 was compared with that of the saline and empty lentivirus treated groups. The expression of TNF-¦Á was significantly reduced compared with the normal saline and empty vector lentivirus treatment groups (P<0.05). Further detection of the expression of Cadherin-11 was also significantly reduced (P<0.05). Conclusion Increased miRNA-144 in the body can attenuate or inhibit the inflammatory response of synovial tissue in arthritis model mice and downregulate the expression of TNF-¦Á, and this effect may be due to the inhibition of Cadherin-11 in synovial cells by miRNA-144. effect. Abstract

Abstract Objective To observe the expression of hypoxia-inducible factor (HIF)-1¦Á in the portal vein of rabbits with schistosomiasis and explore its significance in the activation of smooth muscle cells (SMC). Methods The portal hypertension model was established by infecting rabbits with schistosomiasis cercariae through skin application. Western blot and immunohistochemistry were used to detect the expression levels of HIF1¦Á and type I collagen at 150 days. At the same time, optical microscopy and transmission electron microscopy were used to compare the media thickness and SMC ultrastructure of the portal vein of rabbits in each group. The results were consistent with those of healthy rabbit portal veins. Comparing tissues, the expression of HIF-1¦Á in schistosomiasis rabbits was significantly increased (integrated absorbance was 6.45¡À2.14 and 3.27¡À1.58, respectively, P<0.01), and the content of type I collagen was significantly increased (integrated absorbance was 141.3¡À28.21 and 48.53¡À12.22, respectively. P<0.01). In the model group, portal vein SMC significantly proliferated, transformed into synthetic fibroblasts, and the tunica media thickened (118.29¡À36.52) ¦Ìm. Abstract

? Objective To study the antagonistic effect of cucumber aroma on liver fibrosis in rats. Methods 48 rats were randomly divided into normal control group, model group, normal saline group and cucumber fragrance intervention group. A fully automatic biochemical analyzer was used to detect alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline Phosphatase (ALP) and hematoxylin-eosin (HE) staining were used to observe the microstructure of the liver, and immunohistological staining was performed to detect type I and III collagen, ¦Á-smooth muscle actin (¦Á-SMA) and transforming growth factor ¦Â1 (TGF- ¦Â1). Results The ALT, AST and ALP of the cucumber aroma intervention group were (70.8¡À12.3), (112.3¡À26.4) and (169.1¡À21.2) U/L respectively, which were significantly lower than those of the model group (P<0.01); the cucumber aroma intervention group There was no statistically significant difference in the expression of liver type ¢ñ, ¢ó collagen, ¦Á-SMA and TGF-¦Â1 compared with the normal control (P>0.05). Abstract

Objective To observe the therapeutic effect of Junggar Aconitum and its processed products on type II collagen-induced arthritis (CIA) rats. Methods A blank control group, a model group, a positive control group (dexamethasone acetate tablets 6 mg¡¤kg-1), Junggar Aconitum and its three processed products were established in high, medium and low dose groups. The ELISA method was used to detect the levels of IL-2, SA, and TNF-¦Á in the serum of each group. Pathological sections of the synovial tissue of the knee joint of CIA rats were taken and stained, and the pathological changes of the synovial tissue were observed using an optical microscope. Abstract: Objective To explore the feasibility of constructing a tissue-engineered three-dimensional bronchial model in vitro using human bronchial epithelial cell lines and human fetal lung fibroblast cell lines as seed cells and collagen gel composite Matrigel as scaffold material, in order to study bronchial epithelial cells and Fibroblast interactions provide a model. Abstract

Objective To explore the regulatory effect and possible mechanism of gas molecule hydrogen sulfide (H2S) on collagen synthesis in rat cardiomyocytes. Methods Rat cardiomyocytes H9C2 were randomly divided into 4 groups: normal control group (added to DMEM culture medium), sodium hydrosulfide (NaSH) group (cultured with H2S donor NaHS with a final concentration of 2¡Á10¨C4 mol/L), TGF-¦Â group (culture with a final concentration of 10 ng/ml TGF-¦Â1), TGF-¦Â + NaHS group (culture with DMEM medium containing 2¡Á10¨C4 mol/L NaHS for 30 min, and then add 10 ng/ml TGF-¦Â1 for culture). After 1 hour of culture, Western blotting was used to detect the protein expression levels of p-Smad2 and p-Smad3 in cardiomyocytes. After 72 hours of culture, immunofluorescence staining was used to observe the expression of type I collagen in cardiomyocytes. Results Compared with the normal control group, there was no significant difference in the protein expression levels of p-Smad2 and pSmad3 in cardiomyocytes in the NaHS group, while the protein expression of p-Smad2 and p-Smad3 in cardiomyocytes in the TGF-¦Â group was significantly increased, and the expression of type ¢ñ collagen was significantly enhanced. . Compared with the TGF-¦Â group, the expression of p-Smad2 and p-Smad3 proteins in cardiomyocytes in the TGF-¦Â+NaHS group was significantly reduced, and the expression of type ¢ñ collagen was significantly weakened. Abstract

Objective To preliminarily explore the mechanism of D980-nm semiconductor laser on lipolysis and rejuvenation of SD rat skin, and analyze the differences between different treatment frequencies. Methods 18 clean-grade male SD rats aged 12 to 14 months and weighing 400 to 450 g were used. Taking the rat spine as the dividing line, the left skin was used as the laser irradiation experimental side, and the right skin was used as the normal control side. The experimental animals were randomly divided into 3 groups (n=6). The experimental side (groups A, B, and C respectively) used a D980-nm semiconductor laser in pulse mode with an energy density of 20 J/cm2, a power of 8 W, and a pulse width of 20 ms, pulse frequency 40 Hz, subcutaneous irradiation 1, 2, and 3 times respectively as one course of treatment, with an interval of 5 minutes each time, and an interval of 1 week between each course, for a total of 4 courses. The control side (groups A1, B1, and C1 respectively) was not treated with laser. One week after the end of the fourth course of treatment, the samples were collected and subjected to HE staining and immunohistochemistry staining. The changes in the rat skin tissue structure before and after laser irradiation were observed, the dermal thickness was measured, and fibroblasts and TGF-¦Â1 and bFGF positive cells were counted. Results Compared with groups A1, B1, and C1, the skin tissue structure of rats in groups A, B, and C was significantly improved after D980-nm semiconductor laser irradiation, with a decrease in fat cells, the formation of local small blood vessels, an increase in fibroblasts, and an increase in collagen fibers. The bundles increased, thickened, and were arranged tightly and regularly. The thickness of the dermis, fibroblasts, and TGF-¦Â1 and bFGF-positive cells all increased significantly, and the difference was statistically significant (P<0.05). As the frequency of laser treatment increased, the above indicators gradually increased. Group C was better than groups A and B, and the difference was statistically significant (P<0.05). Conclusion D980-nm semiconductor laser treatment has obvious lipolysis and rejuvenation effect on rat skin, which is closely related to the frequency of laser treatment. Abstract

Objective To study the expression of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in breast ductal carcinoma and their relationship with type IV collagen (collagenIV). Methods Immunohistochemical double staining method was used to detect the expression of MMP-2, MMP-9 and type IV collagen in 60 cases of breast ductal carcinoma and analyze their correlation with tumor size, lymph node metastasis and human epidermal growth factor receptor-2 (C -erbB-2) relationship. Results The positive expression rates of MMP-2 and MMP-9 in breast ductal carcinoma were 75% and 80% respectively, which were significantly higher than 10% in normal breast tissue; MMP-2 and MMP-9 were close to the basement membrane or would break through the basement membrane. The expression of MMP-2 and MMP-9 was significantly enhanced in cancer cells at the membrane. %, 85.1%) expression rate was significantly higher than that of the corresponding control group (58.6%, 65.5%), (64.9%, 67.6%), (46.2%, 53.8%); the expression of type IV collagen was related to MMP-2, MMP- 9 expressions were all negatively correlated. Abstract

Study the selective anti-tumor effect of lidamycin (ldm) and anti-type IV collagenase monoclonal antibody 3g11 immunoconjugates prepared by different coupling methods. Using spdp and smbs as coupling agents, the lysine at position 69 of the LDM prosthetic protein was connected to the 2-iminotetrahydrothiophene-modified monoclonal antibody 3g11 to prepare an immunoconjugate; the immune activity of the conjugate was measured by ELISA. The anti-tumor effects of the two conjugates were observed by cloning formation method and human ht-1080 cell immunodeficiency mouse transplantation model. The conjugate retained the immune activity of monoclonal antibody 3g11 against type IV collagenase, and the killing effect of 3g11-smbs-ldm on HT-1080 cells cultured in vitro was stronger than that of ldm and 3g11-spdp-ldm. Animal experiments show that the tumor inhibition rate of ldm under equimolar dosage conditions is 86.1%, the free ldm group is 71.2%, the 3g11-spdp-ldm group is 77.1%, the inhibitory effect of 3g11-smbs-ldm Significantly enhanced. The average survival time of animals in the 3g11-smbs-ldm group was extended to 163.7%, that of the 3g11-spdp-ldm group was 125.3%, and that of the ldm group was 71.9%. 3g11-smbs-ldm can significantly extend the survival time of tumor-bearing mice. There are significant differences between the two species. The 3g11-smbs-ldm conjugate has more selective anti-tumor effects, significantly improved efficacy, prolonged survival of tumor-bearing animals, and significantly reduced toxicity, and may become a new tumor treatment drug.
