Abstract

Abstract Objective To explore the effects of different replication levels of hepatitis B virus on the progression of liver fibrosis. Methods 173 patients were divided into 6 groups: 3 groups with mild, moderate, and severe chronic hepatitis, and 3 groups with Child-Pugh Class A, B, and C cirrhosis. Radioimmunoassay was used to determine the serum leukocytes of patients with chronic hepatitis B and cirrhosis. Interleukin-6 (IL-6), interleukin-8 (IL-8), type ¢ó procollagen (P¢óP), type ¢ô collagen (c-¢ô) content, and at the same time determine HBV-DVA, according to positive and negative Compare different groups. Results: Serum IL-6, IL-8, P¢óP and c-IV levels were compared when HBV-DNA (+) and HBV-DNA (-), except in the mild hepatitis group and the liver cirrhosis C grade group, there was no significant difference. Abstract

Objective: To explore the establishment of a new method for isolating human fibroblast collagen phagocytic subpopulation (CPSF) and fibroblast non-collagen phagocytic subpopulation (nCPSF), and to screen the differentially expressed genes between the two. Methods: According to the difference in phagocytic functions of CPSF and nCPSF, collagen-coated isothiocyanate fluorescent latex microspheres (COL-FITC-LB) phagocytosis technology and flow cytometry separation technology were used to separate them, and the two were separated through biochip technology and Real-time PCR screens and verifies the differentially expressed genes between the two, and looks for molecular markers that are characteristic of CPSF and nCPSF. Results: CPSF and nCPSF were successfully isolated, and their gene differences were analyzed, and 17 differentially expressed genes were obtained. Compared with nCPSF, 12 genes were up-regulated and 5 genes were down-regulated in CPSF. Three genes that may be related to collagen phagocytosis were verified, namely urokinase plasminogen activator receptor-associated protein (uPARAP), cytochrome b-245, and beta polypeptide (cytochrome b- 245, beta polypeptide, CYBB) and homolog 1 (Hook homolog 1, HOOK1). The expression level of uPARAP in CPSF is 2788 times that of nCPSF; the expression level of CYBB in CPSF is 0.85 times that of nCPSF; the expression level of HOOK1 in CPSF is 2788 times that of nCPSF. 1.96 times, the difference was statistically significant (P<0.05). Conclusion: A new method for separating CPSF and nCPSF was established. The main differentially expressed gene between CPSF and nCPSF is uPARA P, which has an important impact on the collagen phagocytosis function of fibroblasts and is one of the new candidate target molecules for the treatment of collagen metabolism diseases. Abstract

Purpose: Isolate and purify the specific target antigen of anti-glomerular basement membrane (gbm) antibodies, and evaluate its application value in the detection of anti-gbm antibodies. Method: Extract bovine glomeruli from bovine kidneys through screens with different pore sizes , use modified 40g*l-1 sodium deoxycholate to destroy glomerular cells to obtain gbm. After digestion with collagenase, collect the soluble bovine gbm crude antigen. Separate it through monoq anion exchange chromatography column under the condition of ph7.5. The glomerular basement membrane type IV collagen ¦Á chain non-collagen domain 1 [¦Á(IV)nc1] was purified, and its purity and activity were identified by SDS-polyacrylamide gel electrophoresis and westernblot. 2mg*l-1 purified bovine ¦Á(IV)nc1 was coated on an enzyme-labeled plate, and a bovine ¦Á(IV)nc1-elisa method was established to detect anti-gbm antibodies in serum, and a control study was conducted with the classic elisa method using human gbm soluble protein as crude antigen. The serum came from 90 100 patients with known positive anti-human gbm antibodies, 100 normal patients and 50 patients with other kidney diseases. Correlation analysis of the two methods was performed and the sensitivity and specificity of bovine ¦Á(IV)nc1-elisa were calculated. Results: Purification The bovine ¦Á(¢¤)nc1 showed by SDS-polyacrylamide gel electrophoresis as proteins with relative molecular masses of 25¡Á103 and 50¡Á103 and could be recognized by anti-gbm antibody-positive serum; application of bovine ¦Á(¢¤)nc1- The sensitivity and specificity of the ELISA method for detecting anti-gbm antibodies are 100% and 98% respectively, and the correlation with human gbm crude antigen-ELISA is 0.6. Conclusion: The modified sodium deoxycholate method is used to destroy intraglomerular cells. And further separate and purify bovine ¦Á(¢¤)nc1 through monoq ion exchange column to obtain high-purity specific target antigen; the purified bovine ¦Á(¢¤)nc1 can be used as a specific antigen to detect human anti-gbm antibodies.

As the temperature increases, the adsorption amount of acidic yellow G increases, and the adsorption equilibrium conforms to the Langmuir equation; the adsorption kinetic data shows that the adsorption of acidic yellow G increases. Abstract

Objective To observe the effects of dehydrocordine (DHC), the main active ingredient of the traditional Chinese medicine Corydalis Corydalis, on the proliferation and collagen secretion of cardiac fibroblasts (CFs) cultured with high concentration of glucose, so as to provide experimental basis for the clinical application of Corydalis Corydalis. Methods The CFs model cultured in high glucose in vitro was used for experiments. The CFs of suckling mice within 24 hours after birth were isolated by combined digestion with collagenase and trypsin. The cells from the 2nd to 4th generations were cultured in high-concentration glucose (25 mmol/L), and 100, 50, and 25 mg/L DHC were added respectively for intervention. 24, 48 Cell morphology was observed under the microscope after h, cell proliferation was detected by MTT method, cell cycle was detected by flow cytometry, and the content of type I and type III collagen was determined by ELISA.
