Abstract

The DNA sequences, which originated from gene COL1A1, optimized for E.coli codon usage and contained cell adhesion domains of GER units, was amplified by RT-PCR technology from human tissue. The recombinant expression vector pET28a( )-COL encoded the recombinant human -derived collagen (RHDC) was constructed and transformed host E.coli strain BL21(DE3)pLysS. The RHDC production was induced by the addition of IPTG and approximately 40% of the total strain proteins. RHDC could form a triple-helical structure suggested by the SDS-PAGE and Western blot analysis. Cell adhesion assay indicated that RHDC had high cell adhesive character, which makes it suitable for use as biomaterials.

Abstract

Under UV irradiation, using H2O2 as the initiator, collagen/polyvinylpyrrolidone/acrylamide (Collagen/PVP/AM) pH-sensitive hydrogel was prepared by graft copolymerization. The effects of raw material ratio and drying method on gel swelling properties were investigated, and its pH sensitivity and pH swelling-deswelling characteristics were studied. The prepared hydrogel has a fast swelling rate, and the water absorption rate can reach about 94% in 5 minutes. The prepared hydrogel has obvious pH sensitivity and good pH swelling-deswelling reversibility. Fourier transform infrared spectroscopy (FTIR) and differential scanning calorimetry (DSC) results show that while maintaining the collagen triple helix structure, chemical cross-linking occurs between the materials, and the thermal stability of the material is significantly improved, thus expanding the Material application range.

Abstract

Objective To explore the effects of IL-27 and its receptor (wsx-1) on the proliferation, transformation and collagen synthesis of mouse lung fibroblasts induced by TGF-¦Â1. Methods Construction of il-27r/pcdna3.1 overexpression vector. The experiment was divided into 6 groups: normal group, tgf-¦Â1 group, il-27 group, il-27 receptor group, il-27+il-27 receptor group, tgf-¦Â1+il-27+il-27 receptor group body group. The cell growth status was observed under a light microscope, and mtt was used to detect the effect of each treatment group on the proliferation of lung fibroblasts. RT-PCR and western blotting were used to detect the expression of myofibroblast marker a-SMA and types I and III collagen, and immunofluorescence was used to detect the localization and expression of a-SMA protein. Results (1) Both il-27 and il-27r can inhibit the proliferation of lung fibroblasts; (2) tgf-¦Â1 promotes the increase of ¦Á-sma expression in lung fibroblasts and promotes the synthesis of type I and III collagen in lung fibroblasts; (3) il-27 and its receptor can inhibit the expression of ¦Á-sma in lung fibroblasts and reduce the synthesis of type I and III collagen in lung fibroblasts; (4) When il-27/il-27r work together, It has no effect on the proliferation and transformation of lung fibroblasts. Abstract

Porcine whole femur, femoral shaft, and femoral head collagen hydrolyzate were used as samples to study their effects on blood pressure in spontaneously hypertensive rats (SHR). After SHR adaptive feeding for 1 week, they were randomly divided into 11 groups, with 8 animals in each group: the negative control group was gavaged with distilled water, and the 3 samples were divided into 3 dose groups: low, medium, and high, respectively, at 20.0, 40.0, and 60.0 mg/kg. The corresponding collagen hydrolyzate was intragastrically administered at a dose of 10.0 mg/kg, and the positive control group was intragastrically administered captopril at a dose of 10.0 mg/kg. The SHR tail artery systolic pressure of each group was measured every 2 hours after intragastric administration. The results showed that the low-dose femoral head group, whole femur and femoral stem sample groups had no significant antihypertensive effect on SHR, while the mid- and high-dose femoral head groups could significantly reduce the blood pressure of SHR. UV spectrophotometry was used to detect the ACE inhibition rate of the samples. The ACE half-inhibitory mass concentrations (IC50) of the whole femur and femoral head samples were 3.546 and 2.045 mg/mL respectively. It can be seen that femoral collagen hydrolyzate can effectively inhibit the activity of ACE in vitro, but only femoral head collagen hydrolyzate can effectively reduce SHR blood pressure under higher mass concentration conditions.

Abstract

In this study, tilapia bone collagen was used as raw material to prepare active peptides with good calcium chelating ability through enzymatic hydrolysis. Using calcium chelating activity as an indicator, the tilapia bone collagen obtained from the tilapia skeleton after pre-treatment such as meat removal and decalcification was enzymatically hydrolyzed by papain to prepare tilapia bone collagen peptide. Cross-experiment was conducted to optimize the enzymatic hydrolysis process for preparing tilapia collagen chelated peptide. At the same time, the molecular weight and amino acid composition of tilapia bone collagen calcium chelate peptide obtained by enzymatic hydrolysis under optimal conditions were analyzed. The results showed that the optimal enzymatic hydrolysis process conditions were: enzymatic hydrolysis time 4.5 h, pH 6.5, enzymatic hydrolysis temperature 62 ¡ãC, and enzyme bottom ratio 0.6%.
