Abstract In order to reveal the effects of different extraction methods on the structural characteristics and functional properties of plaice skin collagen, fresh plaice skin was used as the research object, and collagen was extracted from it using acid method and enzymatic method to obtain acid-soluble collagen (ASC) and Enzymatically soluble collagen (PSC) was studied, and the structural characteristics and functional properties of these two collagens were comparatively studied. The UV spectrum scanning results showed that both ASC and PSC had strong absorption peaks at 230 nm and no peaks at 280 nm. The obvious absorption peaks are consistent with the structural characteristics of collagen; Fourier transform infrared spectrum scanning results show that both ASC and PSC contain amide A, amide B, amide I, amide II, and amide III, and both maintain the three-dimensional helical structure of collagen. Integrity; SDS-PAGE gel electrophoresis confirmed that both ASC and PSC are type I collagen. The results of hygroscopicity and moisturizing properties showed that with the change of time, the hygroscopicity rates of ASC and PSC showed a gradually increasing trend, while their The moisture retention rate showed a downward trend; the results of turbidity and aggregation characteristics showed that the aggregation kinetic curves of ASC and PSC solutions both showed an S-shaped trend, and the aggregation capacity of ASC was greater than that of PSC during the entire process; water absorption and oil absorption The results show that both ASC and PSC have certain water and oil absorption properties, and the oil absorption properties of both are greater than their respective water absorption properties.
