Abstract

The elemental balance and metabolic balance methods were applied to establish a mathematical model for the expression period of human-like collagen produced by fermentation of recombinant Escherichia coli, and the nonlinear optimization method was used to estimate the unknown parameters in the model. The results show that the model is consistent with the growth and metabolic kinetics model of recombinant E. coli, and the key calculation parameters ¦Áh, ¦Áp and mx of the model are 1.173mol.C-mol-1, 293.814mol.C-mol-1 and 17.878mol respectively. .C-mol-1.h-1. Abstract

Objective To observe the therapeutic effect of Yishen Jubi Pills on rats with type II collagen-induced arthritis (CIA) and its effect on the pathological changes of ankle joint tissue, and to explore the effects of Yishen Jubi Pills on the treatment of rheumatoid arthritis (RA). Vascular endothelial growth factor (VEGF) correlation. Methods: By establishing a CIA rat model, we observed the therapeutic effect of Yishen Guanbi Pills on foot swelling in CIA rats and on the pathological damage of local tissue in the ankle joint. The expression level of VEGF in the synovial tissue was detected by immunohistochemistry. , using gene chip technology to measure the gene expression of VEGF in peripheral blood lymphocytes and liver tissue. Results: The degree of ankle joint swelling of rats in the high, medium and low dose groups of Yishenjuanbi Pills was significantly reduced, and the differences were significant compared with the model group (P<0.05, 0.01, 0.001). Compared with the model group, the high, medium and low dose groups of Yishen Juanbi Pills showed reduced synovial cell proliferation, significantly reduced synovial tissue congestion and edema, reduced vascular proliferation and the number of infiltrating inflammatory cells, significantly reduced pannus formation, and reduced fibrous tissue formation. , the degree of destruction of cartilage and bone tissue is reduced. Compared with the model group, the expression of VEGF in the synovial tissue of the high- and medium-dose Yishenjuanbi Pill groups was significantly lower (P<0.05, 0.01). The expression of VEGF in the synovial tissue of the low-dose group also showed a downward trend. The high and medium dose groups of Yishen Jubi Pills can significantly down-regulate the expression of VEGF-B gene in liver tissue (fold change>1.5). Conclusion Yishen Jubi Pills have a significant therapeutic effect on CIA rats, can significantly reduce the swelling of rat ankle joints, significantly inhibit the pathological changes of CIA rats, and can significantly down-regulate the expression of VEGF in synovial tissue and VEGF- in liver tissue. The expression of B gene can reduce the amount of VEGF in liver tissue and synovial tissue, inhibit the formation of pannus in synovial tissue, and reduce the damage to articular cartilage and bone tissue.

Abstract

Objective To explore the effect of radiofrequency hyperthermia on the expression of type II collagen during the formation of knee osteoarthritis in rabbits. Methods: Fifty-four male rabbits were selected, and their right hind limbs were made into experimental knee osteoarthritis models using the modified Hulth modeling method. After the model was successfully created, the experimental rabbits were randomly divided into the model group, the deer melon polypeptide group and the radiofrequency hyperthermia group. The deer melon polypeptide group was given intramuscular injection of deer melon polypeptide after the model was made. The radiofrequency hyperthermia therapy group was given radiofrequency hyperthermia treatment after the model was made. The model group was not given special treatment after the model was made. On the 7th, 13th and 19th days of treatment, 6 experimental rabbits from each group were sacrificed, and the right medial femoral condyle cartilage was harvested and morphologically scored using the modified Mankins score. At the same time, real-time quantitative PCR technology was used to detect the right side of the experimental rabbits in each group. Type II collagen content of medial femoral condyle cartilage tissue. Results After treatment for the same number of days, it was found that the Mankins score of the right medial femoral condyle cartilage in the model group, the deer melon polypeptide group and the radiofrequency hyperthermia group decreased in sequence, and the type II collagen content of the cartilage tissue increased in sequence. The differences between the groups were statistically significant (P <0.05). As the treatment time in the radiofrequency hyperthermia group prolongs, the Mankins score gradually decreases and the type II collagen content in cartilage tissue gradually increases. The differences between each time point are statistically significant (P<0.05). Conclusion Radiofrequency hyperthermia is significantly more effective than deer melon polypeptide in treating rabbit knee osteoarthritis, and its therapeutic mechanism may be related to the significant increase in type II collagen content in cartilage. Objective To explore the effect of radiofrequency heating on type II collagen expression in a rabbit model of osteoarthritis. Methods Knee osteoarthritis was induced in the right hind legs of 54 male rabbits using modified Hulth modeling. The rabbits were randomly divided into a model group which was not given any special treatment, a Lugua polypeptide group and a radiofrequency hyperthermia group. The Lugua polypeptide group was injected with Lugua polypeptide; the radiofrequency hyperthermia group was treated with radiofrequency irradiation. Six, 12 and 18 days after the treatment, the morphological condition of the rats¡ä right femoral medial condyle cartilages were evaluated using modified Mankins scoring and the type II collagen content of the cartilage was detected using a quantitative PCR technique. Results At the same time points after treatment, the average Mankins scores were decreased in all the 3 groups, with that of the model group was significantly higher than those of both of the other groups, and the radiofrequency hyperthermia group¡äs average score was significantly better than that of the Lugua polypeptide group. The average type II collagen content was significantly increased in all the 3 groups to various extent (the radiofrequency hyperthermia group > Lugua polypeptide group > model group) . For the radiofrequency hyperthermia group, the average Mankins score decreased significantly and the average type II collagen content increased significantly as the treatment continued. Conclusion Radiofrequency hyperthermia is superior to Lugua polypeptide for treating knee osteoarthritis, at least in rabbits. Its therapeutic effectiveness may be related to a significant increase of type II collagen in the cartilage

Abstract

The objective of this study was to develop research of cardiac cells to reestablish 3D tissue architecture in vitro, we performed studies using collagen membrane as three-dimensional scaffold for cardiac cells culture with the principles and methods of tissue engineering. The polymer scaffold provides a 3 -D substrate for cell attachment and tissue formation. Cardiac cells isolated by enzymatic digestion from 1d old neonatal rats were seeded to three-dimensional collagen scaffolds and tissue culture plates. The morphology, beating rate and the metabolic indexes, including specific consumption rate of glucose (q(glu)) , specific production rate of lactate (q(lac)), lactate transform rate ( Y(lac/glu)), specific creatine kinase (CK) and lactate dehydrogenase (LDH) activities of cardiac cells cultured on three -dimensional collagen membrane and tissue culture plates were compared. It was found that cells shape and cells’ CK and LDH activity was no differences between 3D and 2D cultures and cell beat rate on cell culture cluster was slower than those cells cultured on collagen membrane, However the cell glucose consumption and lactate yield rate of cells cultured on cluster was higher than those cells cultured on collagen membrane. After 5 days of cultivation, cardiac cells cultured on collagen membrane scaffolds organized into three-dimensional (3D) aggregates as opposed to the two-dimensional (2D) aggregates mosaic pattern seen in tissue culture plates, and spontaneous and rhythmical contractile 3D cultures in unison were visible to the naked eye and the area of synchronous contract three-dimensional (3D) aggregates reaches 80cm2. The mean value of q(glu), q(lac) and Y(lac/glu) of cultured on three-dimensional collagen scaffold was 7.37 micromol/10(6) cells/d, 2.92 micromol/10(6) cells/ d and 0.38 micromol/ micromol, versus 7.59 micromol/10(6) cells/d, 3.83 micromol/10(6) cells/d and 0.51 micromol/micromol in tissue culture plates. These results demonstrate that cardiac cells immobilized on collagen membrane in 3D cultures maintain similar metabolism activity and contractile function when compared with native cardiac cells. The above results support the idea that engineered cardiac tissue can be used as a model of native tissue for studies of tissue development and function in vitro and eventually for tissue repair in vivo.

Abstract

To improve the stability of small unilamellar vesicles(SUV) in vitro, the permeability of SUV from soybean phosphatidylcholine (PC) coated by collagen and chitosan was studied respectively by using 5(6)-Carboxyflourescein(5(6)-CF) as a fluorescence probe. Results showed that coating with collagen/PC weight ratio of 2 or chitosan/PC weight ratio of 8 decreased significantly the permeability of the liposomal membrane. Moreover, fluorescence polarization method was used to study the influence of coating with collagen and chitosan at the above ratio to PC on the fluidity of the liposomal membrane, by using an intramolecular charge transfer compound 3-methoxy-4′-N, N-dimethylamino flavone derivative(DMMF) as a fluorescence probe. Results showed that the fluidity of the liposomal membrane after coating with the two macromolecules had no obvious changes. It indicates that SUV coated with collagen and chitosan at appropriate weight ratio can significantly improve the stability of the liposomal membrane in vitro without disturbing its fluidity.
