Abstract

We isolated the strain MBL13 with high collagenase productivity from the soil of piled up animal bones.It was identified as Bacillus cereus.We purified and characterized Bacillus cereus collagenase(BCC).The molecular weight of BCC Was 38.0 kDa and the optimum temperature and pH for the enzyme activity were 40¡æ and 8.0 respectively.The enzyme was stable when the temperature was below 50¡æ,but only retained 10%activity when kept at 60¡æ for 1 h.The enzyme activity was stable between pH 7.0-8.5.Some metal ions such as Ca~(2+),Zn~(2+),Mg~(2+) enhanced the enzyme activity, and Cu~(2+) brought the obvious inhibition. In addition, EDTA and EGTA could inhibit the enzyme activity.We suggested that the purified enzyme was a member of the metalloproteases.Based on the experiment of substrate specificity,WC found that the purified enzyme was bone collagenolytic protease,and had a much stronger capacity of hydrolysis fortype ¢ñ collagen than that fortype ¢ò collagen and type ¢ó collagen.By BCC hydrolyzing bone collagen, we obtained polypeptides with different chain lengths.The comparative test indicated that the hydrolysis capacity of BBC was higher than that of standard type ¢ñ collagenase.The results introduced a new swain and a novel collagenolytic protease for industrial enzvme.

Abstract: The macrostructure of the cartilage scaffold was designed using the three-dimensional drawing software Solidworks, and the silk fibroin-type II collagen cartilage scaffold was prepared using 3D printing technology and freeze-drying technology. The density, porosity and elastic modulus of the scaffold were tested experimentally; after chondrocytes were seeded on the scaffold, the proliferation and morphology of the cells on the scaffold were analyzed using three methods: MTT method, HE staining and scanning electron microscopy. The results show that the elastic modulus of the silk fibroin-type II collagen cartilage scaffold is rate-dependent, that is, as the strain rate increases, the elastic modulus of the scaffold increases; the density and porosity of the scaffold are (0.086 6 ¡À 0.008 4) respectively. g/cm3 and (89.3¡À3.26)%. Abstract

Using basa fish skin as raw material, the degreasing process of fish skin by ultrasonic emulsification was studied. Conduct a systematic study on factors such as degreasing agent type, ultrasonic time, degreasing agent concentration, and material-to-liquid ratio during the degreasing process. The degreasing agent selection results show that surfactant NP-10 combined with ultrasonic degreasing effect is better. On this basis, through single factor experiments and orthogonal experiments, it was determined that the optimal process is to select a NP-10 solution with a volume fraction of 1%, ultrasonic treatment for 3 hours under the condition of an ultrasonic power density of 2.22¡Á104W/m3, and the material-to-liquid ratio is 1:5.5(g/mL). Abstract

Objective: To prepare a new type of collagen scaffold material using pangasius skin as raw material, and to detect and analyze the structural characteristics, physical properties and in vitro degradability of the material to explore whether pangasius fish can replace terrestrial mammals as a suitable source of collagen scaffold materials. . Methods Basa fish skin was subjected to repeated degreasing, decolorizing, and impurity removal processes, and then freeze-dried into a film. The crude protein content was measured using the Kjeldahl nitrogen determination method, the structure of the material and the size and distribution of pores were observed and analyzed using a scanning electron microscope, the porosity was measured using the liquid displacement method, and the tensile strength was measured using a universal testing machine. Observe how the viscosity of the material changes with temperature to detect the denaturation temperature of the material. Soak the material in phosphate buffer (0.1 mol/L, pH 7.4) placed in a constant temperature shaking box at 37¡ãC, and detect the water absorption rate and weight loss rate of the material. The crude protein content of the resulting material was 95.2%. The thickness is uniform according to the naked eye. Scanning electron microscopy shows that one side of the material is rough and has a porous structure with varying sizes and even distribution; the other side is smooth and has dense pores. Porosity (55.50¡À1.94)%, thickness (0.66¡À0.10) mm, tensile strength (18.82¡À0.94) MPa. The denaturation temperature of the material without cross-linking is 34¡ãC, and after thermal cross-linking it is 36¡ãC. 48 h water absorption rate (379.77¡À77.81)%. The weight loss rate of the thermally cross-linked material after 28 days was (80.22¡À2.49)%, and the buffer pH was 6.67¡À0.05. Conclusion Basa fish skin collagen scaffold material can be made into biofilms with uniform thickness and different structures of dense layer and loose layer. The material has good mechanical strength and a suitable denaturation temperature, but the degradation rate is fast and needs to be improved. The changes of viscososity with temperatures were detected to determine the denaturation temperature of the material.The material was immersed in the phosphate-buffered solution ( 0.1 mol/L, pH 7.4), which was placed in a constant temperature shaker at 37¡æ, and the water absorption and weight loss rates of the material were detected. Results The crude protein content of the collagen scaffold material was 95.2%, with visually uniform thickness. SEM photographs showed that one side of the material had a rough surface and porous structure, on which varying sizes of pores distributed uniformly; the other side was smooth with dense pores. The porosity of the material was (55.50¡À1.94) %, thickness was (0.66¡À0.10) mm and tensile strength was (18.82¡À0.94) MPa. The denaturation temperature of the material was 34¡æ before thermo-crosslinking and 36¡æ after thermo-crosslinking. The water absorption of the material was ( 379.77¡À77.81)% at 48 h. At 28 d after thermo-crosslinking, the degradation rate was (80.22¡À2.49)%, and the pH value of buffer was 6.67¡À0.05. Abstract

Objective: To observe the effects of total saponins of Thladiantha dubia root (TSTR) on the osteoclast (OC) positive feedback factor leukocytes in blood and knee joint synovium in rats with bovine type II collagen-induced arthritis (CIA). Changes in levels of interleukin-15 (IL-15) and IL-34 to explore the mechanism of action of TSTR in treating CIA in rats. Methods: There were 80 male Wistar rats, 10 of which were randomly selected as the normal group. The remaining rats were injected with an emulsion of bovine type II collagen mixed with complete Freund’s adjuvant into the skin of the tail root and back at multiple points. The same method was performed 7 days later. 14 days after the first injection, the rat arthritis index (AI value) ¡Ý4 was considered as successful modeling. The model rats were randomly divided into model group, TSTR low, medium and high dose groups ( 20, 40, 80 mg¡¤kg-1) and Tripterygium wilfordii polyglycoside (TG)-positive drug group (12 mg¡¤kg-1). Each drug group was given the corresponding drug by gavage for 35 days, once a day. Methods: 60 healthy SD rats were randomly divided into 2 groups, the normal group (10 rats) and the liver fibrosis model group (50 rats). In the modeling group, 40% carbon tetrachloride (CCl4) was intraperitoneally injected to prepare the HF animal model. After successful modeling, they were randomly divided into 5 groups, namely the model group, Eucommia polysaccharide high, medium and low dose groups and Colchicum There are 10 animals in each group in the alkaline group. Samples were collected from the colchicine group (1.0¡Á10-4g¡¤kg-1) and Eucommia polysaccharide high-, medium-, and low-dose groups (0.14, 0.07, 0.035 g¡¤kg-1) after continuous intragastric administration for 8 weeks, respectively. After acid hydrolysis, collagen was separated on a C8 reversed-phase column at a flow rate of 1.0 mL/min using acetonitrile-0.05% trifluoroacetic acid aqueous solution (volume ratio 5:95) as the mobile phase. Using theanine as the internal standard, mass spectrometry was used to determine HYP qualitatively and quantitatively. Selected ion monitoring was performed on m/z132 and m/z175 ions in electrospray positive ion mode. In the range of 11.7 to 117 mg/L, the peak area ratio of HYP to the internal standard theanine and the mass concentration of HYP showed a good linear relationship, with a correlation coefficient of 0.9993. The relative standard deviation of the content determination was 1.87%, and the recovery rate was 97.85% to 101.76%. This method has a simple mobile phase, short analysis time and no need for derivatization. It has strong anti-interference ability and can accurately and quickly determine the HYP content in collagen.

Abstract

Grass carp (Ctenopharyngodon idellus) scales were used as raw materials, and three commonly used industrial proteases were used to enzymatically hydrolyze three fish scales that had been treated with different pretreatment methods. Collagen peptides were prepared through enzymatic hydrolysis, centrifugation, freeze-drying and other processes, and the different pretreatments were compared. The yield and product characteristics of collagen peptides prepared by the method. The results show that the pretreatment method has a significant impact on the nitrogen yield, degree of hydrolysis and amino nitrogen production of the fish scale enzymatic hydrolysis product (P<0.05). The appropriate pretreatment method is heat treatment combined with crushing treatment; the pretreatment method has a significant impact on the product's The influence of molecular weight and distribution is not obvious. The high-performance liquid chromatogram of the product has three major component peaks, which are around 10 000~14 000, 8 000~9 000, and 4 000 ku respectively. The pretreatment method affects the effect of collagen peptides. The DPPH¡¤clearance rate has a significant impact, mainly affected by heat treatment, and the impact of crushing treatment is smaller. Considering the yield and product characteristics of collagen peptides, the ideal fish scale pretreatment method is heat treatment combined with crushing treatment. Under different pretreatment methods of heat, smashing and their combination, the collagen peptide of crass carp scale was obtained after the processes of enzymatic hydrolysis, centrifugalization and freeze drying. The yield and properties of specimens were compared and analyzed. Results showed that the average values of the nitrogen recovery,degree of hydrolysis,amino-N production and anti-oxidant activity ª«in vitroª« were significantly different between different pretreatment groups (P<0.05).The relative molecular weight and distribution did not vary significantly with pretreatment methods.Considering the yield and properties of end-products, the ideal pretreatment method is heat and smashing for collagen peptide extracted from crass carp scale.
