Effects Of Simvastatin On Liver Fibrosis In Rats And Its Mechanism Of Action

Abstract
To observe the effect of simvastatin on liver fibrosis in rats and explore its mechanism. 60 healthy sleep deprivation (SleepDeprivation, SD) rats were randomly divided into control group, model group and drug group. In the model group and drug group, rats were subcutaneously injected with carbon tetrachloride (CCl4) oil solution to create a model. In the drug group, simvastatin (5 mg¡¤kg-1¡¤d-1) was administered by gavage at the same time until the end of the experiment; in the control group, Give an equal amount of olive oil subcutaneously. All animals were sacrificed after 8 weeks. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were detected by a fully automatic biochemical analyzer. Pathological changes in liver tissue were observed by hematoxylin-eosin staining and immunohistochemistry. The expression of type I and III collagen and transforming growth factor-¦Â1 (TGF-¦Â1) in liver tissues of each group was detected. The results showed that compared with the model group, the liver inflammatory reaction and fibrosis of rats in the drug group were milder, the expression of type I and III collagen and TGF-¦Â1 was significantly reduced (P<0.05), and the serum ALT and AST levels were reduced (P< 0.05). It can be concluded that simvastatin can reduce the degree of liver fibrosis, and its mechanism may be related to inhibiting the expression of TGF-¦Â1 and type I and III collagen.
