Abstract
Objective To study the effect of coixenolate on Foxp3+CD4+CD25+ regulatory T cells (Treg) in the peripheral blood of mice with collagen induced arthritis (CIA), and to explore the possibility of coixenolate in the treatment of rheumatoid arthritis. mechanism. Methods Five mice were randomly selected from 25 mice as the normal group, and the remaining 20 mice were used for CIA modeling. After successful modeling, they were randomly divided into a modeling control group of 10 animals and a coixenolate group of 10 animals. The coixolate group was given coixolate injection 25 mL/kg, and the normal group and the modeling control group were injected with normal saline 25 mL/kg, both intraperitoneally once a day. Take the medicine together for 21 days. After medication, arthritis scores in each group were scored, arthritis index was calculated, and flow cytometry was used to determine the proportion of Foxp3+CD4+CD25+ Treg in the peripheral blood of mice. Results Compared with the normal group, the arthritis index of mice in the modeling control group was significantly increased (P<0.01), and the arthritis index of mice in the coixenolate group was significantly decreased compared with the modeling control group (P<0.01). Compared with the normal group, the levels of Foxp3+CD4+CD25+Treg in the modeling control group were significantly decreased (P<0.01). The levels of Foxp3+CD4+CD25+Treg in the coixenolate group were significantly higher than those in the modeling control group after treatment. Conclusion Coixin can increase the proportion of Foxp3+CD4+CD25+Treg in CIA mice and may have a certain immunoregulatory effect in the pathogenesis of CIA.
