Abstract

In order to explore the effect of collagen (HC) and compatibilizer maleic anhydride-grafted low-density polyethylene (LDPE-g-MAH) on the properties of polymer materials, low-density polyethylene (LDPE) was used as the matrix and blended with HC/LDPE composite materials and HC/LDPE-MAH composite materials were prepared by extrusion method, and the composite materials were injection molded into splines of different specifications. The effects of HC and LDPE-g-MAH content on the structure and properties of HC/LDPE and HC/LDPE-MAH composites were studied through characterization methods such as mechanical property testing, SEM and thermal analysis. The results show that when the HC addition amount is 5wt%, the tensile strength of the HC/LDPE composite material reaches the maximum value of 15.824MPa; the addition of LDPE-g-MAH can significantly improve the interfacial adhesion, improve the material’s mechanical properties and thermal stability. When the HC content is 20wt% and the LDPE-g-MAH content is 4wt%, the tensile properties of the HC/LDPE-MAH composite material are optimal.

Abstract

Objective To explore the anti-inflammatory effect and mechanism of gingerol on bovine type II collagen-induced arthritis (Collagen Induced Arthritis, CIA) rats. Methods Six-week-old female Wistar rats were randomly divided into blank control group, model group, gingerol group and tripterygium glycosides group, with 10 rats in each group. A CIA model was prepared to observe the effects of gingerol on joint pathology and arthritis index in CIA rats, as well as its effects on rat serum tumor necrosis factor-¦Á (TNF-¦Á), interleukin-1 (IL-1), and interleukin-1¦Â (IL- 1¦Â) expression. Results Gingerol can inhibit the swelling of ankle joints in CIA rats, reduce arthritis index, reduce serum TNF-¦Á, IL-1 and IL-1¦Â levels, and reduce joint synovial cell proliferation, synovial layer and subsynovial layer lymphocytes. Infiltration, its efficacy was equivalent to that of the Tripterygium wilfordii polyglycoside group (P>0.05). Conclusion Gingerol has a therapeutic effect on arthritis in CIA rats, and its mechanism may be related to the down-regulation of TNF-¦Á, IL-1 and IL-1¦Â.

Abstract

The full-length cDNA sequence of the ¦Á1 gene (col1a1) encoding grass carp muscle type I collagen was cloned for the first time using PCR and race methods. It is 5772 bp, and its open reading frame is 4347 bp, encoding 1448 amino acids. The results of blast homology analysis showed that the amino acid sequence of grass carp col1a1 gene has high homology with zebrafish and goldfish, 93.90% and 93.60% respectively, showing high conservation. Phylogenetic tree analysis shows that this gene is in the same branch as zebrafish and goldfish, and has the closest relationship. Bioinformatics analysis shows that the relative molecular mass of grass carp col1a1 protein is 137.2ku, the theoretical isoelectric point is 5.44, and it is an ¦Á-helix ¦Â-sheet triple helix structure protein. There are 18 triple helical repeat domains, 22 low-complexity domains, and 17 functional domains. The col1a1 protein has two calcium-binding regions 33~69 and 1249~1355 and one zinc-binding region 62~104. The tissue expression results detected by the semi-quantitative RT-PCR method showed that the col1a1 gene was expressed in eight tissues of grass carp muscle, intestine, hepatopancreas, gills, skin, fin rays, kidneys and spleen, among which the col1a1 gene was expressed in the skin, gills The mRNA expression levels in , kidney, and fin ray tissues were higher than those in the other four tissues (p<0.05). Abstract

Bromelain and alcalase enzymes were used to perform composite enzymatic hydrolysis of Nile tilapia skin collagen. The in vitro antioxidant activity of the enzymatic hydrolyzate was studied. Experiments show that the enzymatic hydrolysis product has strong superoxide anion/hydroxyl radical scavenging activity and reducing ability. Ultrafiltration membranes with different molecular weight cutoffs are used to separate the enzymatic hydrolyzate into five components, namely tgh-¢¡ (>10ku), tgh-¢¢ (10¡«5ku), tgh-¢£ (5¡«3ku), tgh-¢¤ (3¡«1ku) and tgh-¢¥ (10ku). Among them, the tgh-¢¥ component showed the strongest superoxide anion radical scavenging activity, so gel filtration, ion exchange and reverse high-pressure liquid chromatography were used to further separate and purify this component. The purified peptide has strong superoxide anion radical scavenging activity, and its ic50 value is 4.6 ¦Ìg¡¤ml-1. Mass spectrometry analysis showed that the molecular weight of the peptide ranged from 311.3 to 932.8u.

Abstract

Collagen fibers were modified with ferric iron as a carrier, and catalase was fixed on the carrier through glutaraldehyde cross-linking. The prepared immobilized catalase protein had a loading capacity of 16.7 mg/g and an enzyme activity yield of 35%. This paper examines the surface tension and surface adsorption properties of an aqueous solution of undenatured collagen biosurfactant (CBS). The results show that the surface tension of CBS aqueous solution decreases with the increase of CBS concentration, and finally levels off, reaching 55.92 mN/m. Near the isoelectric point, the static charge on the CBS surface decreases, significantly reducing its surface tension. At low temperatures, the surface activity of CBS solution is better. When the temperature continues to rise (>35¡ãC), the surface tension of CBS solution increases sharply due to denaturation. The adsorption of CBS aqueous solution belongs to Langmiur type adsorption. According to the ¦Ã-lgC, ¦£-C and C/¦£-C fitting curves, the saturated adsorption amounts are similar, which are 1.86¡Á10^-10, 1.97¡Á10^-10 and 2.03 respectively. ¡Á10^-10 mol/cm^2.
