Abstract

This article successfully prepared bovine Achilles tendon type I collagen fibers using an acid-enzyme combination method. Its secondary structure is consistent with the structural characteristics of type I collagen. AFM results found that the collagen fibers were uniform in thickness, had a D periodic striated structure, and a diameter of 135 nm. ¡À0.20 nm, evenly dispersed in the solution and showing a porous micro-network structure; its tensile strength and elongation at break reach 1.293MPa and 93.77% respectively; its hydrophobic properties are improved compared to pure type I collagen, and the contact angle is 68.9¡ã, the water absorption rate, water retention rate and water vapor permeability are 37.13%, 19.44% and 2749.77 g/(m2/d) respectively; its porosity is 89.54%, and the average cumulative adsorption and cumulative desorption specific surface areas are 8.4251 m2 respectively. /g and 12.8046m2/g, showing the characteristics of large pore volume, small specific surface area and large pore size. Analysis of the microstructure and physical and chemical properties of bovine Achilles tendon type I collagen fibers provides a new and reliable theoretical basis for the deep processing, shaping and application of natural collagen fibers.

Abstract

[] Objective To explore the effect of Smad7 gene on the expression of type I and type III collagen (ColI and ColIII) in rat glomerular mesangial cells (MsC), and to provide experimental basis for trying to use Smad7 for gene therapy in renal fibrosis. Methods: The recombinant expression plasmid containing Smad7 was transfected into rat MsC through liposome mediation, and G418 screening and Northernblot and Westernblot methods were used to identify the expression changes of transfected positive MsC clones Col¢ñ and Col¢ó using RT-PCR and Westernblot methods respectively. . Results The positive MsC clones (S-22 and S-26) that stably and highly expressed Smad7 were successfully established, and it was confirmed that the expression of Col¢ñ and Col¢ómRNA and protein of the two positive MsC clones were significantly inhibited, and the expression of Col¢ñ and Col¢ómRNA of S-22 clone was reduced respectively. 47% and 56%, and its protein expression decreased by 65% and 54% respectively. Conclusion Smad7 may reduce the progression of renal fibrosis by inhibiting the production of Col¢ñ and Col¢ó in tissues.

Abstract

Objective To study the effect of infrared rays on the expression of c-Jun and type ¢ñ and type ¢ó collagen in fibroblasts (HSF) cultured in vitro, and to provide a theoretical basis for the molecular mechanism of photoaging damage caused by infrared rays. Methods: Primary skin fibroblasts were extracted and cultured. The fibroblasts were divided into control group (without infrared irradiation) and experimental group (infrared irradiation). MTT was used to detect cell activity in each group. Real-time quantitative PCR and immunocytochemistry (ICC) were used. Methods The expressions of c-Jun and type ¢ñ and ¢ó collagen mRNA and protein in each group were detected. Results MTT test showed that compared with the control group, infrared irradiation in each group of the experimental group had varying degrees of inhibitory effects on the proliferation of HSF; infrared irradiation down-regulated the expression of type ¢ñ collagen mRNA and protein. As the irradiation dose increased, the expression of type ¢ñ collagen decreased. The expression of mRNA and protein decreased more obviously (P<0.01); after 12 hours of infrared irradiation of fibroblasts, the expression of type III collagen mRNA and protein showed a dose-dependent decrease (P<0.05, P<0.01). The expression of type collagen mRNA and protein was up-regulated in a dose-dependent manner (P<0.05, P<0.01); infrared irradiation up-regulated the expression levels of c-Jun mRNA and protein in a dose-dependent manner as the irradiation dose increased. (P<0.05, P<0.01). Conclusion Infrared irradiation of human skin fibroblasts increases the expression of c-Jun, inhibits the expression of type ¢ñ collagen, and interferes with the expression of type ¢ó collagen. Abstract

Type I collagen extracted from tilapia skin was mixed with different concentrations of phycocyanin, and a composite hydrogel was prepared using the self-assembly properties of collagen. The air permeability, micromorphology, and infrared spectrum of the composite hydrogel were investigated. , swelling rate and antioxidant properties. The results showed that the air permeability of the composite hydrogel increased significantly. As the phycocyanin content increased, the micromorphological pores and swelling properties of the hydrogel increased. The infrared spectrum showed that the preparation method did not destroy the characteristic absorption peaks of the two proteins. In the antioxidant experiment, the composite hydrogel has DPPH free radical and hydroxyl radical scavenging ability, showing excellent antioxidant ability.

Abstract

In order to study the effect of high-density CO2 (dense phase carbon dioxide, DPCD) pre-treatment on the extraction rate of collagen from salamander skin, freeze-dried salamander skin was used as raw material, DPCD was used to pre-treat salamander skin, and collagen from salamander skin was extracted by enzymatic method. A single factor experiment and a response surface experiment were designed to optimize the process conditions of DPCD treatment of salamander skin using the collagen extraction rate as an evaluation index. The results show that the optimal temperature for DPCD treatment of salamander skin is 32¡ãC, pressure 30 MPa, and time 6 h; under these processing conditions, the model predicts that the extraction rate of collagen from salamander skin is 39.70%, and the verification value is (39.48¡À0.74)% , there is no significant difference (P>0.05); compared with the collagen extraction rate of the raw material without DPCD treatment ((20.63¡À0.46)%), the extraction rate increased by 18.85%; the microstructure shows that the salamander skin after DPCD treatment The tissue is loose, the regular arrangement of micellar fibers is destroyed, and it appears porous; the electrophoresis pattern shows that after salamander skin is treated with DPCD, the collagen maintains the typical structural characteristics of type I collagen. DPCD treatment of salamander skin significantly improves the collagen extraction rate while maintaining the structural characteristics of collagen in salamander skin.

Abstract

In order to make full use of the collagen protein in fish bones from fish fillet processing waste, single factor and orthogonal experimental methods were used to study the optimal process for extracting tilapia bone collagen protein by pepsin under acidic medium conditions. The refined collagen protein was SDS-PAGE electrophoresis, ultraviolet and Fourier transform infrared scanning, circular dichroism detection, amino acid analysis, thermal denaturation temperature measurement and other methods are used to analyze the physical and chemical properties of collagen proteins. The results show that: the pH of the lactic acid solution is 2.6, the temperature is 30¡ãC, the material-to-liquid ratio is 1:30 (g/mL), the amount of enzyme added is 200 U/g, and the extraction time is 24 h. The extraction rate of crude collagen protein under this process condition reaches 70.28%. , the yield is 12.76%; tilapia bone collagen protein contains 2 ¦Á chains and 1 ¦Â chain, which is a typical type I collagen protein; the main amino acids of tilapia bone collagen protein are glycine, proline, and hydroxyproline Acid, glutamic acid and alanine; the thermal denaturation temperature of tilapia bone collagen protein is 34.5 ¡æ. Studies have shown that using pepsin to extract fish bone collagen protein has higher extraction rate and purity. Fish bone,containing plenty of collagen,is the main offal from fish fillet processing. The collagen from Tilapia bone after removing impurities was extracted by pepsin digestion in acid medium condition. The effects of extraction medium,temperature,material to liquid ratio,pepsin addition ,extraction duration on yield of collagen were investigated. The optimum conditions using lactic acid as medium to extract collagen were pH 2.6,material to liquid ratio(w/v) 1:30,30 ¡æ,200 U/g pepsin addition,extraction time 24 h,and the yield of extracted collagen reached 70.28%(12.76% when compared with the initial weight of Tilapia bone). Then, the extracted crude collagen was further purified and its physicochemical property was determined by the methods of SDS-PAGE,UV and FT-IR scanning,DSC,CD and amino acid analysis. The results indicated that the extracted Tilapia bone collagen belongs to typical collagen¢ñwith two ¦Á and one ¦Â chains. Furthermore, the Tilapia bone collagen contains high percentages of Gly,Pro,Hyp, Glu and Ala and its denaturation temperature was 34.5 ¡æ. Abstract

This article used layer-by-layer self-assembly technology to alternately assemble oppositely charged heparin and type IV collagen onto the surface of the material, and studied the effects on endothelial progenitor cells before and after assembly and with different numbers of assembly layers. After titanium is activated by alkali, hydroxyl groups are added to the surface, which combines with the carboxyl groups in polylysine to form a positively charged amino surface on the material. Then, heparin and type IV collagen with heterogeneous charges are alternately added to the material. Multiple self-assembled layers are formed on the surface. Fourier transform infrared grazing was used to detect changes in the surface groups of the material after each step of treatment, toluidine blue was used to quantitatively characterize the amount of heparin assembly on the surface of the material, atomic force microscopy was used to observe the surface morphology changes of the material before and after assembly, and the assembly process was tracked by measuring the water contact angle.
