Abstract
This experiment uses catfish skin as raw material to compare the physical and chemical properties of collagen extracted by alkali method, salt method, hot water method, acid method and enzymatic method. By analyzing the amino acid composition, ultraviolet spectrum and Fourier transform of different collagens, Research on infrared spectroscopy, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), thermal denaturation temperature, scanning electron microscopy images and rheological properties, from molecular structure to microstructure Comprehensive analysis and comparison of collagen. The results show that the UV spectra and infrared spectra of collagen extracted by the five methods are relatively similar and are typical collagens; the thermal denaturation temperatures of collagen extracted by alkali method, hot water method, salt method, acid method and enzymatic method are respectively 63.21, 71.70, 49.49, 29.8, 22 ¡æ; SDS-PAGE results show that the collagen extracted by the acid method, enzymatic method, and hot water method all belong to type I collagen, and the protein structure is well retained; high performance liquid chromatography It shows that the collagen extracted by the alkali method and the salt method is in the form of polypeptides; through rheological properties research, it is found that the collagen gel extracted by the hot water method has the highest stability. It can be seen that different extraction methods have different effects on the amino acid composition, rheological properties, thermal denaturation temperature and microstructure of collagen.
