Optimization Of Decolorization Process Of Squid (Ommastrephes Bartrami) Skin Collagen Extract Using Response Surface Methodology

Abstract
The activated carbon adsorption method was used to remove pigments from the squid (Ommastrephes bartrami) skin collagen extract, and the decolorization conditions were optimized to further utilize the squid skin collagen. Under the condition that the characteristic absorption wavelength of the pigment was determined to be 420 nm, the effects of activated carbon addition amount, decolorization time, solution pH value and decolorization temperature on the pigment removal effect of squid skin collagen liquid were studied. On the basis of the single-factor experiment, the decolorization time was selected to be 30 minutes, and a three-factor and three-level Box-Behnken central combination response surface analysis test of activated carbon addition amount, solution pH value, and decolorization temperature was conducted. The results show that the optimal conditions for activated carbon to adsorb pigments from squid skin collagen liquid are 4.2% activated carbon addition, solution pH 2.27, and decolorization temperature 35¡ãC. Under these conditions, the decolorization rate of squid skin collagen liquid is 45.6%, and protein and amino acid losses are observed. The rates are 19.65% and 22.93% respectively. After decolorization, squid skin collagen has the highest glycine content (136.02 mg/g, relative content 23.05%), glutamic acid, arginine and proline content, while the total cysteine and methionine content is less than 1 %, consistent with the characteristics of aquatic protein amino acids and type I collagen amino acids.
