Abstract
The extraction and physical and chemical properties of collagen from sea cucumber (stichopus japonicus) were studied. The results show that low-temperature extraction can obtain acid-soluble collagen (PSC) that is dissolved by pepsin and removes telopeptides. The maximum absorption peak of psc measured by UV-visible scanning is located at 220nm; amino acid analysis found that among 1000 total amino acid residues, there are 329 glycine and 66 hydroxyproline. The ratio of hydroxyproline to proline is 0.69, the content of tyrosine, phenylalanine and histidine is low, and it may not contain cystine. These characteristics are consistent with the characteristics of type I collagen. The thermal stability temperature (ts) of sea cucumber pseudo-sea cucumber PSC measured by differential calorimetric scanning (DSC) is 57¡ãC, which is 5¡ãC lower than bovine type I collagen; the total sugar of PSC was measured by phenol-sulfuric acid and methylene blue methods. and mucopolysaccharide content are 0.61% and 0.48% respectively; after purification by deae52, pure protein with the mucopolysaccharide removed can be obtained. The sds-page shows that the pseudo-sea cucumber PSC extracted in this study does not contain impurity proteins. The main components are collagen ¦Â and ¦Á chains, with a small amount of ¦Ã chain; the ¦Á chain is similar to the ¦Á1 chain of vertebrate type I collagen, and the molecular composition of PSC is (¦Á1)3.
