Abstract
Grass carp scales were used as raw materials to extract acid-soluble collagen (asc) and enzyme-soluble collagen (psc) from the fish scales, focusing on related research including thermal stability, in vitro enzymatic degradability and collagen sponge material properties. , and compared with porcine skin collagen (pc) of mammalian origin. Experimental results show that the three types of collagen prepared are typical type I collagen and have a complete triple helix structure; the thermal denaturation temperature of pc (41.6¡ãC) is significantly higher than asc (34.8¡ãC) and psc (35.2¡ãC); The in vitro enzymatic degradation performance of the three types of collagen is affected by the type of hydrolase, collagen extraction method, collagen source, collagen heating history, and the degree of self-assembly of the protein. Collagenase, trypsin and papain all have varying degrees of degradation ability for freshwater fish collagen, but collagenase has the strongest degradation ability. Under the same conditions, the in vitro enzymatic degradation rates of the three collagens were asc>psc>pc; The in vitro enzymatic degradation rate of collagen significantly increased after thermal denaturation treatment, while the in vitro enzymatic degradation rate decreased to varying degrees after in vitro self-assembly treatment; the collagen sponge materials obtained after freeze-drying of the three collagen samples have different mechanical properties and Tissue structure, asc and psc sponges are porous but weak tensile strength sponge materials, while pc is the opposite.
