Abstract
¡¾¡¿Objective to observe the effect of exogenous human b-type natriuretic peptide on the expression of cardiomyocyte apoptosis factors and type I and III collagen in rats with chronic heart failure. Methods 30 chronic heart failure model rats were randomly divided into a recombinant adenovirus group carrying human b-type natriuretic peptide gene (ad-hbnp group, 14 rats), a recombinant blank adenovirus group (ad-track group, 8 rats), The normal saline group (ns group, 8 animals) were given intraperitoneal injection of human b-type natriuretic peptide gene recombinant adenovirus, recombinant blank adenovirus and 1 ml of normal saline, once a week for a total of 4 weeks. Another 10 rats that were not modeled but underwent sham surgery were set as the sham operation group; the rats were sacrificed after 4 weeks, and RT-PCR was performed to detect bcl-2, bax, caspase-3 and type I and III collagen mRNA in myocardial tissue. content. Results After ad-hbnp treatment, the expression of bcl-2 mRNA in the myocardial tissue of rats in the ad-hbnp group was higher than that in the ad-track group and ns group; the expression of baxmRNA was higher than that in the sham operation group; the expression of caspase-3 mRNA was higher than that in the ad-track group and ns group. group and sham operation group were increased. The expression of type ¢¡ and ¢£ collagen mRNA in the ad-hbnp group was lower than that in the ad-track group. Conclusion Intermittent administration of ad-hbnp can affect the expression of apoptosis-related genes in cardiomyocytes in rats with chronic heart failure and reduce the expression of type I and III collagen in the interstitium of cardiomyocytes. ?
