Changes In Gene Expression In Liver Cells Of Rats With Collagen-Induced Arthritis

Abstract
Objective To explore the pathogenesis of rheumatoid arthritis by studying the gene expression profile of liver tissue in rats with collagen-induced arthritis. Methods The lower bound fold change method of dChip (Dec.2009 version) analysis software was used to analyze genes with expression differences of more than 1.5 times. The chip type was GeneChip Rat Genome230, the gene library was Affymetrix Gene, the reference was the GenBank gene library, and the functional pathways were referred to the KEGG database. The rat whole gene expression profiling chip was used to study the gene expression profiles of collagen-induced arthritis and normal rat liver tissue, and compare the gene expression differences in the liver tissue of model rats and normal rats. Results The results showed that there were 1,009 differentially expressed genes in rats with collagen-induced arthritis, including 480 up-regulated genes and 529 down-regulated genes. Differentially expressed genes are mainly involved in biological process regulation, stimulus response, cell communication, negative regulation of cell cycle, immune response, stress response, angiogenesis, endothelial cell differentiation and other functions. The metabolic and signaling pathways involved mainly include apoptosis pathway, calcium regulation pathway, TGF-¦Â pathway, coagulation function pathway, inflammatory response pathway and other pathways. The genes that were differentially expressed in the model group mainly included Aldh1a7, Bad, Gdf10, Ccar2, Slc1a3, Il1b, Il7, Vegfb, IgG-2a, etc., and the genes that were differentially expressed were mainly Ugt2b, Mx2, Usp18, Comt, Zfp354a, Oas1a, and G1p2. , Irf7, etc. Conclusion Collagen-induced arthritis is a systemic immune disease involving abnormal expression of multiple genes. The differentially expressed genes screened initially reflect the pathogenesis of rheumatoid arthritis and provide important clues for the prevention and treatment of rheumatoid arthritis.
