Abstract
Using yak bone as raw material, collagen was extracted using acid method and enzymatic method respectively, and its physical and chemical properties were analyzed. The results were analyzed through amino acid composition, ultraviolet spectrum, Fourier transform infrared spectrum, heat shrinkage temperature, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and scanning electron microscope image analysis. Collagen for comparison. The results show that: the extracts obtained by the two methods are all typical collagen, with the maximum UV absorption peak appearing at around 230 nm wavelength; acid-soluble collagen (ASC) and enzymatic-soluble collagen (pepsin-soluble) The heat shrinkage temperatures of collagen (PSC) are 40.12?¡æ and 40.94?¡æ respectively, and the denaturation enthalpy values are 0.25?J/g and 0.22?J/g respectively. Infrared spectroscopy and SDS-PAGE analysis show that ASC and PSC are mainly composed of ¦Á It is composed of three subunit components: ¦Â, ¦Ã, ¦Â, and ¦Ã. It belongs to type I collagen and has a complete triple helical structure. Scanning electron microscopy results show that the collagen extracted by the two methods retains a relatively complete fiber network structure, but The collagen structure extracted by the acid method is relatively evenly distributed. Taken together, there is not much difference in the physical and chemical properties of the collagen extracted by the two methods, but the collagen extracted by the enzymatic method has a higher yield, and the acid extraction method has a lower cost.
