Abstract
Type II collagen (CII) has been proven to be the most important autoimmune antigen in rheumatoid arthritis (RA). The use of oral chicken type II collagen (CCII) to induce immune tolerance to treat RA marks one of the major advances in RA treatment strategies. .Based on the successful cloning of chicken type II collagen gene (CCOL2A1) internationally, the scientificity and feasibility of using pcDNA-CCOL2A1 gene vaccine strategy to treat RA were discussed. The cloned CCOL2A1 gene was connected to the eukaryotic expression vector pcDNA3. 1(+) is the gene vaccine pcDNA-CCOL2A1. The expression vector pcDNA-CCOL2A1 was then transfected into COS-7 cells. Westernblot analysis showed that pcDNA-CCOL2A1 expresses CCII in a secreted form in COS-7 cells. The ¦Á subunit of the polypeptide chain. Subsequently, CCII-induced Wistar rats were used as the RA model, that is, CIA, to systematically observe the therapeutic effect of one-time tail vein injection of three different doses of pcDNA-CCOL2A1 gene vaccine of 20, 200, and 400 ¦Ìg/kg on CIA arthritis. Results It showed that on the 5th day after injection, the joint swelling of rats in the 200 ¦Ìg/kg group was significantly reduced, and there was a significant difference compared with the empty vehicle group (P<0.05), while the two-dose groups of 20 and 400 ¦Ìg/kg were significantly different from the empty vehicle group. No therapeutic effect was observed in the group comparison (P>0.05). Six weeks after injection, the anti-CII antibody concentration level and TNF-¦Á concentration in the serum of CIA rats were significantly lower in the 200 ¦Ìg/kg treatment group than in the empty vehicle group (P<0.05) , while the 400 ¦Ìg/kg group increased significantly, while the 20 ¦Ìg/kg group showed no significant change. In addition, the concentration levels of cytokines TGF-¦Â and IL-10 increased significantly in the 200 ¦Ìg/kg group compared with the empty vehicle group (P<0.05 ), while there was no significant difference between the 20 and 400 ¦Ìg/kg groups. The splenocyte lymphocyte proliferation experiment showed that the spleen cells of the rats in the 200 ¦Ìg/kg dose pcDNA-CCOL2A1 gene vaccine treatment group were significantly less responsive to CII than the empty vector group. (P<0.05). This result shows that the 200 ¦Ìg/kg dose of pcDNA-CCOL2A1 gene vaccine can significantly inhibit the joint swelling of CIA rats, reduce the levels of anti-CII antibodies and TNF-¦Á, and increase the levels of cytokines TGF-¦Â and IL-10. , reduces the proliferation response ability of spleen cells, and has a significant therapeutic effect on arthritis in CIA rats.
