Abstract
Abstract Objective To study the changes in collagen expression in atrial tissue in patients with rheumatic valvular heart disease and atrial fibrillation (AF), and to explore the mechanism of atrial fibrosis in patients with atrial fibrillation and its role in the occurrence and persistence of atrial fibrillation. Methods: 75 cases of rheumatic heart disease Patients with valvular disease who underwent valve replacement surgery were divided into 3 groups, including 34 cases in the sinus rhythm group, 11 cases in the paroxysmal atrial fibrillation group, and 30 cases in the chronic atrial fibrillation group. About 100 mg of right atrial appendage tissue was obtained during the operation, and bitter taste was used. Acid sirius red staining method was used to analyze the amount and distribution of collagen deposition in atrial tissue, and semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) method was used to determine the mRNA levels of type ¢ñ collagen and type ¢ó collagen in atrial tissue. .Results (1) There is significant fibrosis in the atrial tissue of patients with atrial fibrillation, and the degree of fibrosis is positively correlated with the inner diameter of the left atrium (r=0.390, P=0.001) and the duration of atrial fibrillation (r=0.320, P=0.005); (2 ) Compared with the sinus rhythm group, the expression of type ¢ñ collagen mRNA in the atrial tissue of patients with paroxysmal atrial fibrillation (P£¼0.05) and chronic atrial fibrillation (P£¼0.001) was significantly increased; the expression of type ¢ó collagen in patients with atrial fibrillation was significantly increased. Although the mRNA expression of type I collagen increased, the difference was not statistically significant. The mRNA expression level of type I collagen was positively correlated with the inner diameter of the left atrium (r=0.336, P=0.004) and the duration of atrial fibrillation (r=0.339, P=0.003). .Conclusion There is obvious fibrosis in the atrial tissue of patients with atrial fibrillation. The molecular remodeling of type I collagen may be one of the molecular mechanisms of atrial fibrosis in atrial fibrillation and is related to the occurrence and persistence of atrial fibrillation.
