Abstract
Caco-2 cell monolayers were cultured after the 6-well transport plate was coated with collagen. The transmembrane resistance of the Caco-2 cell monolayer and the positive drug propranolol were measured under different collagen types, rat tail collagen gel concentrations and gel spreading times. The apparent permeability coefficients of passive diffusion of Er and atenolol were determined, and it was found that different collagen coating conditions had no significant effect on the integrity and transport capacity of Caco-2 cell monolayers. Therefore, a stable collagen-coated 6-well transfer plate system for Caco-2 cell culture can be constructed by spreading 2 ¦Ìg¡¤cm-2 rat tail collagen for 1 hour, providing an in vitro research technology method for studying the absorption of the material basis of traditional Chinese medicine compounds.
