Abstract
Study the thermal degradation process of tilapia skin collagen fiber in hydrogen peroxide solution system, and the changes in the amino acid composition, molecular weight and properties of the degradation products in the reaction system. The results showed that within 1 hour of reaction, very few collagen degradation products were released into the solution, which was not enough to test the gel strength and viscosity of the sample. After reacting for 1 to 3 hours, the apparent viscosity of the solution in the reaction system increases. The reaction solution can condense into a gel after cooling. The gel strength gradually increases with time, indicating that the number of collagen molecules released into the aqueous solution increases and the solute mass concentration increases. improve. After 3 to 5 hours of reaction, the apparent viscosity of the reaction solution decreased with time, the solute mass concentration in the reaction solution increased, and the gel strength decreased, indicating that this stage is dominated by the cleavage of peptide chains of procollagen molecules, resulting in further degradation of collagen. The electrophoresis results showed that in the sample reacted for 4 h, the proportion of bands with molecular weights below 130 kD increased, indicating that the ¦Á chain fragments in the sample increased. Analysis of the amino acid composition showed that the amino acid composition of the degradation product was slightly different from that of fish skin, but there was no significant change in the amino acids of the gelatin product at different reaction time points, indicating that the synergistic thermal effect of hydrogen peroxide was mainly achieved by hydrolyzing peptide bonds to achieve collagen fiber orientation.
