Detection Of Collagen Content In Different Tissues Of Crucian Carp Using High-Performance Liquid Chromatography

Abstract
In order to improve the utilization rate of aquatic product collagen, pre-column derivatization-high performance liquid chromatography was used to detect the hydroxyproline content in crucian carp skin, fish meat, fish scales and swim bladder tissue, and then the hydroxyproline content was converted into Corresponding collagen content. Crucian carp samples were hydrolyzed with hydrochloric acid at 110¡ãC for 18 h, derivatized with dansyl chloride, and detected using high-performance liquid chromatography-ultraviolet method. Chromatographic column: reversed-phase C18 column (250 mm¡Á4.6 mm, 5 ¦Ìm); mobile phase: A is methanol, B is tetrahydrofuran-methanol-0.05 mol/L sodium acetate (1:15:84, V/V) solution, Gradient elution was used; flow rate was 1 mL/min; column temperature was 40¡ãC; detection wavelength was 254 nm. The results show that hydroxyproline has a good relationship in the range of 20 to 400 ¦Ìg/mL (R2=0.999 8), the lowest detection limit is 0.18 ¦Ìg/g, and the average recovery rate at different levels is 80.22% to 83.33%, relatively The standard deviation range is 0.31%~1.84%. Analysis of actual samples showed that the collagen contents in fish skin, fish meat, fish scales, and fish bladder tissues were 106.62, 24.75, 166.94, and 115.05 mg/g respectively. The analysis method established in this experiment is simple, fast, has good sensitivity and reliability, and can be used for the quantitative analysis of collagen in different tissues of aquatic products.
