Differences In The Expression Of Collagen And Matrix Metalloproteinases In The Left And Right Atria Of Patients With Chronic Atrial Fibrillation

Abstract

This article aims to study whether there are differences in the mRNA levels of collagen, matrix metalloproteinases (MMPs), and tissue inhibitors of metalloproteinases (TIMPs) in the left and right atria of patients with chronic atrial fibrillation. A total of 45 patients were selected to undergo thoracotomy for heart valve replacement, including 27 patients with chronic atrial fibrillation and 18 patients with sinus rhythm. During the operation, left and right atrial appendage tissues were harvested and type I collagen was measured using semi-quantitative RT-PCR. , type III collagen, matrix metalloproteinase-1,9 (MMP1, MMP9) and matrix metalloproteinase inhibitor-1 (TMP1) mRNA expression levels. The results showed: (1) Compared with the sinus rhythm group, the mRNA expression levels of type I collagen, MMP1, and MMP9 in the left and right atrial myocardium of patients in the chronic atrial fibrillation group were all increased (P<0.05), and the expression levels of TMP1 were all increased. Down-regulated (P<0.01); (2) There was no statistically significant difference in the mRNA expression levels of type I collagen, type III collagen, MMP1, MMP9, and TMP1 between the left and right atrial myocardium groups of patients in the sinus rhythm group (P >0.05); (3) The expression level of MMP1 mRNA in the right atrial muscle tissue of patients in the chronic atrial fibrillation group was higher than that of the left atrium (P<0.05), and the expression level of MMP9 mRNA in the left atrial muscle tissue was higher than that of the right atrium (P <0.01); (4) Regardless of the left atrium or the right atrium, the mRNA expression levels of MMP1 and MMP9 were positively correlated with the mRNA expression levels of type I collagen and negatively correlated with the mRNA expression levels of TMP1. The above results indicate that the imbalance of MMP1/TMP1 and MMP9/TMP1 mRNA expression levels in atrial myocardial tissue causes changes in type I collagen transcription levels, which may be the molecular basis of atrial myocardial interstitial fibrosis in patients with chronic atrial fibrillation.
