Dual Effect Of Mechanical Strain On The Pelvic Fibroblast Cells

Abstract
Objective: To explore the bidirectional effects of mechanical force on pelvic floor fibroblasts. Methods: The sacral cardinal ligament tissues from patients who underwent total vaginal hysterectomy for non-pelvic organ prolapse in our hospital from January 2013 to January 2015 were collected, primary cultured fibroblasts were grouped as follows according to different mechanical force loading parameters : (1) No force control group; (2) 1mm 4h0.5Hz group; (3) 4mm 4h0.5Hz group. CCK-8 method was used to detect the cell proliferation activity of each group after intensive intervention, and Western Blot was used to detect the expression of type ¢ñ and ¢ó collagen in the cells of each group. Results: After loading human pelvic floor fibroblasts with different mechanical parameters, compared with the control group, the cell proliferation activity of the 1 mm 4h0.5 Hz group was enhanced (188.22% ¡À 10.53%), and the cell proliferation activity of the 4 mm 4h0.5 Hz group was weakened. (66.09%¡À4.06%), the differences were statistically significant (P<0.05). Western Blot detected changes in the expression levels of collagen types I and III in each group. Compared with the control group, the expression of collagen types I and III in human pelvic floor fibroblasts in the 1mm 4h0.5 Hz group was significantly increased (P<0.05 , P<0.001), while the expression of type I collagen in human pelvic floor fibroblasts in the 4mm 4h0.5Hz group was significantly down-regulated (P<0.001, P<0.05). Conclusion: Mechanical force has a bidirectional regulatory effect on pelvic floor tissue damage or repair. The mechanism may be that mechanical force has a bidirectional regulatory effect on pelvic floor fibroblasts to promote proliferation or damage, and bidirectionally regulates the metabolism of type ¢ñ and ¢ó collagen.
