Effect And Mechanism Of Total Glucosides Of Paeony On Synovial Cells In Rats With Collagen Arthritis

Abstract
Objective To study the effect and mechanism of total glycosides of paeony (tgp) on synovial cells in collagen-induced arthritis (cia) rats. Methods Chicken type II collagen was used to induce rat cia model. Rat synovial cells were isolated and cultured by collagenase and trypsin digestion. The ultrastructural changes of synovial cells were observed with transmission electron microscope. The proliferation ability of synovial cells was detected by MTT method. The IL-1 activity in the membrane cell culture supernatant was measured using the mouse thymocyte proliferation method, and the tnf¦Á and pge2 contents were measured using the radioimmunoassay. Results TGP can effectively improve the ultrastructural changes of synovial cells in cia rats, inhibit their excessive proliferation response and the levels of IL-1, tnf¦Á and pge2. Conclusion TGP has a significant inhibitory effect on hyperfunctional synovial cells in cia rats, and its mechanism of action may be related to its ability to inhibit the excessive proliferation and secretion of synoviocytes.
