Abstract
Objective: To explore the protective effect and mechanism of Compound Huanglian Capsule (CRCC) on renal lesions in rats with diabetic nephropathy (DN). Methods: Streptozotocin (STZ) was used to replicate the early DN rat model, and the animals were divided into normal group, model group, CRCC high, medium and low dose groups (the crude drug content was 4.36, 2.18, 1.09g.kg-1 respectively ), Xiaoke Pill (XKW) 0.80g.kg-1 treatment group, intragastric administration, once a day, 30 days later, fasting blood glucose (FBG), urea nitrogen (BUN), creatinine (Cr), insulin (Ins) were detected and urine protein (Upro); the morphological changes of renal tissue were observed under light microscope; the expression of transforming growth factor ¦Â1 (TGF-¦Â11) and type IV collagen (IV-C) protein in glomerular mesangial cells was detected by immunohistochemistry. Results: CRCC can reduce the levels of FBG, BUN, Cr and Upro in rats, increase Ins, reduce the expression of TGF-¦Â1 and IV-C protein in glomerular mesangial cells, and improve the morphological abnormalities of renal tissue. Conclusion: CRCC has a protective effect on renal microcirculatory lesions in early-stage DN rats and delays the progression of chronic DN pathology. This effect may be related to its inhibition of TGF-¦Â1 and IV-C protein expression in renal tissue.
