Effect Of Erxian Chubi Decoction On Collagen-Induced Arthritis In Rats

Abstract
Objective To explore the effect and mechanism of Erxian Chubi Decoction on collagen-induced arthritis (CIA) in rats. Methods Healthy Wistar rats were randomly divided into normal group, model group and Erxian Chubi Decoction low and high dose groups. Rat CIA models were established in all groups except the normal group. The Erxian Chubi Decoction low-dose and high-dose groups were given Erxian Chubi Decoction 8 and 16 g/kg respectively by gavage from the 7th day after modeling, once a day for 4 consecutive weeks. Evaluate the incidence rate and clinical scores, observe the pathological morphology and radiological changes of joint tissues, and detect the pro-inflammatory cytokines interleukin (IL)-1¦Â, IL-6, and tumor necrosis factor (TNF)- in the joint tissue homogenate supernatant and serum. Changes in the content of ¦Á and serum inflammatory mediator prostaglandin (PG) E2. Results: The incidence of CIA in rats in the model group was 100%, and the highest clinical score of arthritis was 8.2. The ankle joints showed obvious histopathological changes such as inflammatory cell infiltration, pannus formation, cartilage and bone destruction, as well as joint space narrowing or widening, and articular surfaces. Imaging changes such as moth-eaten damage, IL-1¦Â, TNF-¦Á, IL-6 and serum PGE2 levels in joints and serum were significantly increased. Compared with the model group, the Erxian Chubi Decoction low-dose and high-dose groups can significantly reduce the incidence of arthritis (60%, 40%, respectively), clinical scores (5.7, 3.8, respectively) and improve joint histopathological changes. , the high-dose Erxian Chubi Decoction group can also significantly inhibit the degree of bone destruction and reduce the levels of IL-1¦Â, TNF-¦Á, IL-6 and PGE2 in joints and serum. The low-dose Erxian Chubi Decoction group also significantly inhibits Contents of IL-1¦Â, IL-6 in serum and IL-6 in joints. Conclusion Erxian Chubi Decoction can control the occurrence and development of CIA in rats, and its mechanism may be related to the effective inhibition of systemic and local pro-inflammatory cytokines and inflammatory mediators in the system and joints.
