Abstract
Objective: To explore the effect of the ethanol extract of Malus truncatula on the proliferation of fibroblasts and the production of type ¢ñ collagen in patients with progressive systemic sclerosis (PSS) and normal humans. Methods: The MTT colorimetric method was used to observe the effect of the extract of Malus truncatula on the proliferation activity of fibroblasts and its antioxidant effect, the effect of the extract on cell apoptosis was detected by flow cytometry, and the effect of the extract on PSS was detected by RT-PCR and Westernblot. Effects of type ¢ñ collagen mRNA expression and protein synthesis in patients. Results: Malucarpus extract can inhibit the proliferation, type I collagen mRNA expression and protein synthesis of fibroblasts in patients with PSS in a dose- and time-dependent manner, but has no cytotoxic or apoptosis-inducing effects. The extract has no effect on the proliferation and collagen synthesis of normal human fibroblasts, but has a certain protective effect on fibroblast damage caused by hydrogen peroxide. Conclusion: The ethanol extract of Malus truncatula can inhibit the proliferation of fibroblasts and the synthesis of type ¢ñ collagen in patients with PSS, and has antioxidant effects.
